Aim:For clearly sensitive or resistance to fluconazole intracellular content of fluconazole in Candida albicans, and whether by intracellular content of fluconazole in Candida albicans to fluconazole changes of different fungi play a role of different antifungal therapy.Methods:This study established the use of liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS) determination of intracellular content of fluconazole in Candida albicans. Through the use of LC-MS/MS method for fluconazole (FLC) resistance of Candida albicans strain,100strain and103international standard strain SC5314strain for48h intracellular content of fluconazole monitoring. Through the micro-liquid dilution method for detection of36clinical Candida albicans strains its drug sensitivity to fluconazole, and through the LC-MS/MS method for determination of various strains of intracellular content of fluconazole. Also, through the use of LC-MS/MS method, can accurate clinical strains of intracellular content of berberine hydrochloride were determined.Result:First, LC-MS/MS method for determination of intracellular content of fluconazole in Candida albicans method, can accurate dialogue candida intracellular to determination of fluconazole. Seconded, candida albicans intracellular content of fluconazole with the extension of time and gradually increase, and at24-36h reached the maximum number of intracellular drug concentration. Third, Candida albicans to fluconazole susceptibility is related to its intracellular content of fluconazole, and Candida albicans bacteria sensitive intracellular content of fluconazole is significantly higher than resistant bacteria intracellular content of fluconazole. Forth, in fluconazole and berberine hydrochloride in under the synergistic antifungal mechanism, fluconazole can increase the berberine hydrochloride into intracellular content of Candida albicans.Conclusion:To sum up, in the fluconazole use antifungal role played by, Candida albicans intracellular content of fluconazole and its sensitivity has a certain link; Fluconazole in combination with other drugs of antifungal effect, Candida albicans fluconazole of intracellular content both increase and decrease, or remain unchanged. Said understand Candida intracellular content of fluconazole change is not the primary reason for create synergies, there may be other targets or the change of regulation pathways and play a role of synergistic antifungal; In fluconazole and berberine hydrochloride used in play a role of synergistic antifungal, berberine hydrochloride in Candida albicans increase intracellular content is one of the main reasons for its play a role of synergistic antifungal, but its mechanism is not entirely dependent on the content of berberine hydrochloride into intracellular. Because of fluconazole can increase the berberine hydrochloride into intracellular content of Candida albicans and indirectly promote the fluconazole antifungal role into full play.
