Pharmacodynamic Experimental Study On Canarium Album Vinegar Treatment Of Pharyngitis And Discussion On Its Mechanism

Objective:Research on the pharmacodynamics and acute toxicity of Canarium album vinegar pharyngitis treatment from folk recipes includes canarium album vinegar antibacterial, anti-inflammatory, antipyretic, analgesic and eliminating phlegm etc., acute toxicity test, chemical composition analysis and explore its mechanism of treatment of pharyngitis, which in order to make the preliminary evaluation of its efficacy, safety and function of the material basis and provide experimental basis for Canarium album vinegar further study.Method:Taking Canarium album as raw material to obtain Canarium album vinegar by traditional fermenting method, then the content of potential anti-inflammatory compounds in Canarium album vinegar:phenolic was determined. To research the effect and mechanism of Canarium album vinegar, we will randomly divide experimental animals into five groups:1. Canarium album vinegar high dose group;2. Canarium album vinegar middle dose group;3. Canarium album vinegar low dose group;4. Positive control group;5. Model control group. We observe the anti-inflammatory, analgesic, antipyretic and anti-dryness effect of Canarium album vinegar through the following tests:(1) antimicrobial test in vitro;(2) antibacterial experiment in vivo;(3) the inhibition of capillary permeability method to test the permeability of absorbance value of the abdominal cavity blood capillary of mice by Canarium album vinegar’s influence;(4) the inhibition of inflammatory swelling method to test the inhibition ratio of foot plantar swelling caused by formaldehyde in mice by Canarium album vinegar’s influence;(5) the cotton ball implantation in mice method to test the dry weight of granuloma of mice by Canarium album vinegar’s influence;(6) analgesic test:hot plate method and torsion body method to test the average pain threshold and the changes of the mice body writhing times, calculate the pain inhibition rate;(7) test the amount of phenol red shedding in mice’s windpipe. The study not only research on the pharmacodynamics of Canarium album vinegar pharyngitis treatment, but we also built in vitro inflammatory model by LPS-stimulated murine RAW264.7macrophages to study anti-inflammatory activity of Canarium album vinegar and discussed the potential anti-inflammatory mechanism. Result:(1) Based on ultraviolet absorption spectrum, the content of polyphenol in Canarium album vinegar is15.51mg/g and larger than others fruits;(2) Canarium album vinegar has a strong antibacterial effect against the Escherichia coil, stapgylococcus aureus, and pseudomonas aeruginosa;(3) Canarium album vinegar has somewhat protection for mice from causing death by the stapgylococcus aureus and pserdomonas aeruginosa.(4) The high dose group and middle dose group have obvious restraining effects on raising the abdominal cavity capillary permeability of mice (p<0.05).(5) The high dose group and middle dose group both could strikingly restrain the intestins swelling caused by formaldehyde (p<0.05, p<0.01). And the effect could be last more than24h.(6) The high dose group and the middle dose group both could obviously control the formation of the mice’s granuloma in the cotton ball (p<0.05, p<0.01).(7) The high dose group (15ml/kg) have a remarkable controlling effect in1h,4h,6h,8h compared with the blank control group, it has a more distinguishing discrepancy (p<0.01, p<0.05), and the middle dose group and the iow dose group (7.5ml/kg,3.75ml/kg) have a dropping tendency for controlling pyrexia, but it doesn’t unveil any obvious discrepancy.(8) The mice with Canarium album vigenar high and middle dose groups showed fewer writhes of abdominal cavity, and increased the pain threshold in the hot-plate test.(9) The excretion of phenol red in mice in Canarium album vinegar low group tends to be increasing, high and middle concentration can raise obviously the excretion of phenol red.(10) Canarium album vinegar has a rather strong restraining effect on the inflammatory cells RAW264.7, moreover, the best concentration was the middle dose (15mg/ml).Conclusion:Canarium album vinegar has a better anti-inflammation, antibacterial, antipyretic, analgesia effect, which is the pharmacological basis for the treatment of pharyngitis. Canarium album vinegar has a rather strong restraining effect on the inflammatory cells RAW264.7, which may be one of the mechanisms of healing pharyngitis.
