Objective:From the relationship between the formation of acute soft tissue injury after swelling and aquaporin-depth studying, it is expected to clarify the regulatory mechanism of curing-injury cataplasma promoting acute soft tissue injury after the swelling which was subsided. The research of the way of cell aquaporin controlling acute soft tissue injury will be able to provide new drug targets, and may find a new method of treating tissue swelling.Methods:Totally54SD rats were randomly divided into three groups: normal control group, modeling group, drug-treated group. After establishing the model of soft tissue swelling with acute injury, the rats were killed respectively in six time phase points, namely1h,6h,1d,3d,5d, and7d,three in each group at each point. In marked area some tissue was put out, and in the taken tissue,the proportion of dry and wet method was used for the detection of the water content of skeletal muscle,and the Western Blot and qPCR method for the AQP-1、AQP-3protein and the level of gene expression, then correlation analysis was made.Results:1. the determination of moisture content in skeletal muscle: after modeling, the moisture content in skeletal muscle both model group and the drug-treated group showed a hump-like changes,the3day up to the highest, then gradually decreased. Model group at each time point after modeling muscle tissue water content with the drug-treated group compared with the normal control group increased (P<0.05), but the former two at1h,6h,1d three time points without see a significant difference (P>0.05), the drug-treated muscle tissue water content in the modeling3d,5d and7d were significantly lower than the same point in time the model group (P<0.01).2. Determination of AQP-1gene and protein expression in skeletal muscle:AQP-1mRNA expression levels of the drug-treated group in the modeling1d higher than the normal control group, the difference was statistically significant (P<0.05).3d,5d,7d three time points, the drug-treated group, model group AQP-1mRNA expression levels were significantly higher than the normal control group (P<0.01), while the drug-treated group compared with the model group, the difference was significant statistical significance (P<0.01). AQP-1protein expression at any time point trends and their gene expression consistent trend in each group skeletal muscle.3.Determination of AQP-3gene and protein expression in skeletal muscle:model group and drug treated skeletal muscle AQP-3mRNA expression trend was also found hump-like changes, the highest in the third day of the expression, and then gradually decreased.3d,5d,7d three time points, the drug-treated group, model group AQP-3mRNA expression levels were significantly higher than the normal control group (P<0.01), while the drug-treated group compared with the model group, the difference was significant statistical significance (P<0.01). Groups of skeletal muscle in AQP-3protein expression at any point trend and the trend of gene expression.Conclusion:1. In normal skeletal muscle tissue, AQP-1, AQP-3expression stability.2. AQP-1, AQP-3expression trends with athological changes in the process was time phase changes in skeletal muscle with traumatic soft tissue swelling.3. The curing-injury cataplasma could affect the expression of AQP-1, AQP-3in the skeletal muscle of rats traumatic soft tissue swelling, relieve soft tissue swelling with acute injury,and accelerate the process of repair after the injury...
