Objective:This study underwent detrusor retrograde tracer to observe the lumbosacral spinal cord slice,clear the sacral parasympathetic nucleus(SPN) position, accurate cause of super-sacral spinal cord injury and sacral spinal cord injury in experimental animal models. Observation of the different segments of the sacral nerve root if there are positive fibers, thereby positioning the need to cut off the sacral nerve roots, to avoid any loss of feeling and reflection in sacral segments.Lay the foundation for subsequent experiments.Methods:the rat bladder wall injection of WGA-HRP, continuing to survive for48hours after WGA-HRP injection in rats. Then perfusion-fixed,drawing,sliced,histochemical reaction and microscope.According to the results of retrograde tracing,clear the sacral low center-the location of the urinary bladder detrusor and the marks of bone,to ensure that the injured segments select the correct. At the same time clear the main gathering area of the rat bladder afferent nerves.Followed by the sacral spinal cord injury and sacral spinal cord injury andsacral spinal cord injury on the joint cut off the main bladder afferent nerve to observe the morphology of rat bladder and urodynamic changes.Results:WGA-HRP injected into the bladder wall retrograde tracing, histological,TMB method coloration observed under the microscope,the results show:the distribution of bladder primary afferent neurons in the spinal cord spinal cord L6lateral horn of the Ministry to S1(L6common)visible HRP neurons in the spinal cord L6and S1segments the Ⅶ layer lateral gray matter of the lateral border of the lateral band, also known as the sacral parasympathetic nucleus (SPN).Primary afferent neurons of the rat bladder cell bodies of dorsal root ganglion(DRG),mainly in the L6-S1segments of DRG distribution of L1-S3segments.Sacral spinal cord injury group detrusor leak point pressure joint cut off the main bladder afferent neurons were significantly higher than the sacral cord injury group and sacral spinal cord injury, the difference was statistically significant(P<0.01),sacral injury group,the sacral spinal cord injury in the joint cut off the main bladder afferent group detrusor leak point pressure than the control group,the sacral spinal cord injury group was significantly decreased,the difference was statistically significant(P<0.01);sacral spinal cord injury group, bladder capacity, bladder compliance than the sacral cord injury group and the sacral spinal cord injury jointly cut the bladder afferent nerve was significantly reduced, the difference was statistically significant(P<0.01),the sacral spinal cord injury group,sacral spinal cord injury on the joint cut off the main bladder afferent nerve group, bladder capacity,bladder compliance than the control group significantly increased sacral spinal cord injury group,the difference was statistically significant(P<0.01).Conclusion:According to the experimental results, clearly the position clear to the urinary bladder detrusor low center the sacral parasympatheticnucleus to accurately resulting in the sacral spinal cord injury and sacral spinal cord injury in experimental animal models.Clear the main part of the bladder afferent nerve positioning of the need to cut off the sacral nerve roots,to the maximum extent possible to avoid any loss of sacral segments of feeling and reflection.And according to theurodynamic results verify itsfeasibility.Laid the foundation for the subsequent experiments.
