| Streptomyces sp. R-527F was isolated from the marine sediment samples collected from high latitude Arctic region. It showed strong antibacterial activity against S.aureus and B.subtilis.Several macrolides were isolated from the fermentation broth of the strain Streptomyces sp. R-527F by bio-guided isolation. The biological activities of compounds P1-P3were studied. Furthermore, the preliminary study of fermentation and scale-up of strain Streptomyces sp. R-527F was conducted.The fungal strain Penicillium sp. C-5with higher β3-glucosidase activity was isolated from the runoff sediment samples collected from Ny-Alesund,Arctic. The culture broth of strain C-5showed antifungal activity. So we tried to investigate the bioactive natural products,and several diterpenoid were isolaed.The fermentation broth of Streptomyces sp. R-527F was extracted with petroleum ether and ether acetate successively.The organic layer was evaporated to dryness to afford crude extract,which was separated on silica gel column chromatography.Then the fractions obtained were further separated by chromatography on Sephadex LH-20and preparative HPLC. The structures were determined on the basis of MS, H-NMR and C-NMR spectroscopic analyses and comparison with the literatures. Four compounds(P1-P4) were isolated from petroleum ether fraction and three compounds(P5,P7and P8) from ether acetate fraction. Compound P7was isolated as natural compound for the first time and P8was a novel compound. Using the methods described,five compounds(A-2-1, B-2-1, B-2-2, B-2-3and B-2-4) were isolated from the fermentation broth of Penicillium sp. C-5.Compounds P1-P3showed antibacterial, antitumor and immunosuppressive activity,but to varying degrees. Preliminary study on the fermentation of Streptomyces sp. R-527F demonstrated it had stronger antibacterial activity when the artificial sea water of the medium contained5%NaCl orly.In addition,we succeeded in scaling-up the fermentation of the strain C-5from shake flask to5L bioreactor. |
