Objectives:To observe the hypoxia-induced HU-VEC in the glutathione peroxidase (GSH-PX) activity and lactate dehydrogenase (LDH) leakage volume changes, study ginkgo biloba extract/tea polyphenols and combination groups’impact on HU-VEC in the hypoxia environment.Methods:To detect the endothelial cell cytotoxicity of Ginkgo biloba extract/tea polyphenols and simvastatin trough the MTT method, in order to determine drug concentrations of Ginkgo biloba/tea polyphenols and simvastatin in subsequent experiments. Put endothelial cells in the hypoxia enviroment, by measuring each treatment group’s leakage of LDH and activity of GSH-PX at each time point, study protection effects of Ginkgo biloba extract and tea polyphenols on endothelial cells under hypoxic environment.Results:(1) According to the drug toxicity experiment, we find EGB groups have no cytotoxicity in100μg/ml range; but TP groups show cell growth inhibition in100μg/ml range; cell growth rates of combination groups are close to the negative group, there is no significant difference between combination groups and the negative group; simvastatin groups have no cell growth inhibition in200μg/ml range. So,we selected the EGB100μg/ml、TP25μg/ml and EGB25μg/ml with TP25μg/ml as follow-up experimental drug concentrations;(2)Endothelial cells are damaged in the hypoxia environment, the HU-VEC cells’LDH leakage is increased but the GSH-PX activity is decreased,the extent of cell damage has relationships with hypoxia time, with the extension of hypoxia time, the extent of damage gets worse.(3)EGB、TP and the combination of the two can reduce endothelial cells’LDH leakage and elevate cells’GSH-PX activity to some extent in the hypoxia environment,it’s mean that Ginkgo biloba extract and tea polyphenols can protect endothelial cells which in the hypoxic environment. But with hypoxia time extends, the protective effect falls. In the hypoxic period, the combination-groups’effects are better than Ginkgo biloba extract alone group and tea polyphenols alone group.
