Objective:Acute upper respiratory infection is a common respiratory infectious disease. We have known that, the upper respiratory tract mucosal immunity dysfunction was one of the most important reasons of acute upper respiratory infection repeated attacks. The present clinical treatment measures are difficult to solve the problem. In traditional Chinese medicine view, acute upper respiratory infection falls under the category of exterior syndrome according to the clinical symptoms of it. The methods of relieving the exterior syndrome with of pungent-warm drugs, pungent-cool drugs and strengthening body resistance drugs show good therapeutic effect, but the in-depth function and mechanism are unclear. This thesis research started from the perspective of adjusting the function of upper respiratory tract mucosal immunity system. The mice model of upper respiratory tract mucosal immunity dysfunction was established by cold stimulation. In order to observe the characteristics of the therapeutic effect and to reveal the scientific connotation of the exterior-despelling decoctions in the treatment of acute upper respiratory infection, mice were given with p-hemolytic streptococcus bacteria through nasal and abdominal based on upper respiratory tract mucosal immunity dysfunction. In addition, the antibacterial experiments of the exterior-despelling decoctions were performed.Methods:1. The effect of exterior-despelling decoctions on the mouse model of upper respiratory tract mucosal immunity dysfunctionThe mice were stimulated by the cold environment of-20℃. The optimal stimulated time would be determined by the mouse survival rate and the slgA content and lysozyme activity in the saliva of the mouse. The effect of exterior-despelling decoctions on the model would be evaluated by slgA and lysozyme activity in saliva of the mice, ELISA and turbidimetric method would be chosen.2. The effect of exterior-despelling decoctions on the mice model of upper respiratory tract mucosal immunity dysfunction with β-hemolytic streptococcus bacteria intranasal inoculation.The mouse model of upper respiratory tract mucosal immunity dysfunction was established_by15minutes of-20℃cold stimulation and inoculated intranasally with β-hemolytic streptococcus bacteria. The optimal bacterial concentration and the time of collection of saliva samples would be determined by the mice survival rate and lysozyme activity in the saliva of the mice at different time points. The effect of exterior-despelling decoctions on the model would be evaluated by slgA and lysozyme activity in saliva of the mice, ELISA and turbidimetric method would be chosen.3. The effect of exterior-despelling decoctions on the mouse model of upper respiratory tract mucosal immunity dysfunction with β-hemolytic streptococcus bacteria intraperitoneal injectionThe mouse model of upper respiratory tract mucosal immune dysfunction was established.And then the mice were injected intraperitoneal of β-hemolytic streptococcus bacteria. The effects of exterior-despelling decoctions on mouse model were observed.4. Antibacterial effects of exterior-despelling decoctions in vitroThe antibacterial activities of exterior-despelling decoctions in vitro were observed. Result:1. The model group was placed in the cold environment of-20℃for15minutes, which made the slgA and lysozyme activity in the mice saliva lower than the control group. The mouse model of upper respiratory tract mucosal immunity dysfunction was established. Pungent-warm drugs could improve the slgA content and lysozyme activity of mouse model.2. The mice were placed in the cold environment of-20℃for15min and nasal infected with200μ1concentration of5×107CFU/ml β-hemolytic streptococcus bacteria. The lysozyme activities in the saliva of the mice were significantly lower than the control group after four hours. Pungent-warm drugs could improve the sIgA content of the mice model.3. Exterior-despelling decoctions had no protective effect on the mice model of upper respiratory tract mucosal immune dysfunction with β-hemolytic streptococcus bacteria intraperitoneal injection.4. Exterior-despelling decoctions had no antibacterial ability on β-hemolytic streptococcus bacteria in vitro.Conclusion:1. This study has established the mice model of upper respiratory mucosal immune dysfunction successful.2. Pungent-warm drugs can prevent and cure of the acute upper respiratory infection by improving the function of upper respiratory mucosal immune system.
