| ObjectiveTo investigate the affection of proliferation, apoptosis and related genechanges on Non-Small Cell Lung Cancer cell line (A549) caused byHyperthermia and Etoposide.MethodsConventional cultivation of A549cells in the1640culture medium with10%fetal bovine serum. The cell were divided randomly into four groups: controlgroup, hyperthermia treatment group, etoposide treatment group and combinedhyperthermia and etoposide therapy group. The cell proliferation were detectedwith MTT test and the cell apoptosis were detected with flow cytometry and theRNA of the cells were abstracted and underwent RT-PCR to test the mRNAexpression of Hsp70, Bcl-2and Bax(compared with the β-actin).ResultsBy hyperthermia, etoposide and combined treatment on A549, whichdecreased cell survival. MTT test showed that except48h hyperthermia groupwith the control group was not statistically significant, Absorbance values ofother groups compared with control group were lower (P<0.05).Flow cytometryshowed the apoptosis rate of hyperthermia group, the etoposide group andcombined treatment group compared with the control group significantlyincreased (P<0.01), The expression level of Bcl-2and Hsp70is lower than thecontrol group (P<0.01) while the level of the Bax was higher than the controlgroup(P<0.01).and the highest rate of apoptosis rate in the combination group. ConclusionHyperthermia, etoposide and combined therapy could reduce the non-smallcell lung cancer cell proliferation significantly. The effects of combined treatmentgroup was stronger than that of cisplatin alone. Hyperthermia, etoposide andcombined therapy can induce apoptosis in A549cells, combined therapyinduced apoptosis was stronger than alone. combined therapy inducedapoptosis was stronger than alone. Meanwhile its expression level of Hsp70and Bcl-2decreased, Bax expression levels increased. |
