ObjectiveThe aim of the study was to evaluate the effect of Tanshinone ⅡA on theosteogenic differentiation of rat bone marrow mesenchymal stem cells(BMSCs)MethodsVitro extraction and amplification bone marrow mesenchymal stem cellsfrom SD rats were proliferated to passage three and then induced to osteogenicdifferentiation in conditional culture medium.We added Tanshinone ⅡA withdifferent concentrations(5×10-2μg/ml,5×10-3μg/ml and5×10-4μg/ml)into theexperimental groups and culture medium into the blank groups. MTT automatedcolormetric microassay was used to test the proliferation rate.The osteogenicdifferentiation associated genes,such as ALP,COLL-I,BMP-2and Runx2,wereinvestigated by RT-PCR.We observed the morphology and the mineralization ofthe cells by HE staining and Alizarin red staining.Results1、The proliferation rate of P3BMSCs was increased significantly inexperimental groups when compared with the blank groups (P<0.05). It wasconcentration dependent.2、The expression of the osteogenic differentiation associated genes(ALP,COLL-Ⅰ, BMP-2and Runx2)was increased (P<0.05) in experimental groups. 3、The first96h HE staining of the experimental group and control groupcan be found in the experimental group BMSCs density, showing the typicalwoven-like BMSCs of control group is relatively sparse.4、The Alizarin red staining was positive in experimental groups and theblank groups did not occur.ConclusionsTanshinone Ⅱ A could stimulate the proliferation rate and osteogenicdifferentiation of BMSCs.
