ObjectiveThrough the detection of14-3-3σ, P73α gene and protein expression inbreast different histologic features, to explore both the role of breast cancerdevelopment and their relationship.MethodsImmunohistochemical P-V method and semi-quantitative RT-PCR methodwere detected in94cases of invasive breast cancer (45cases by RT-PCR),15cases of ductal carcinoma in situ,15cases of atypical ductal epithelialhyperplasia,15cases of catheterpapilloma,20cases of breast adenosis, and20cases of breast cancer, next to the the5cm organization of14-3-3σ, p73αprotein and mRNA expression, and retrospective analysis of both breast cancerclinical pathological parameters of the relationship and bothrelevance.Results1, The expression of14-3-3σ protein in breast lesions and its relationshipwith clinicopathological parameters in invasive breast cancer: invasive group,ductal carcinoma in situ group, atypical hyperplasia, papilloma, adenosis group,next to the cancer positive rate of5cm group were29.8%,53.3%,73.3%,86.7%,80.0%,90.0%, the positive rate of invasive cervical cancer group wassignificantly lower than other groups, the difference was statistically significant(all P <0.05). Atypical ductal epithelial hyperplasia, intraductal papilloma, breastadenosis group paraneoplastic5cm difference was not statistically significant (P >0.05). Invasive breast cancer in14-3-3σ protein expression and histologicalgrading, TNM stage, HER-2receptor and axillary lymph node metastasis.2, p73α protein expression and its relationship with clinicopathologicalparameters in invasive breast cancer: p73α protein in the invasive group,carcinoma in situ group, atypical hyperplasia, papilloma, adenosis groupparaneoplastic5cm in positive rates were77.7%,53.3%,40.0%,33.3%,10%,0,invasive group was significantly higher than other groups, the difference wasstatistically significant (P <0.05). p73α protein and breast cancer TNM stage,ER and PR receptors and axillary lymph node metastasis.3, the mRNA levels of breast lesions,14-3-3σ and P73α gene expressionwith clinicopathological parameters in invasive breast cancer: invasive breastcarcinoma in14-3-3σ mRNA in the relative expression levels (0.216±0.042)was significantly lower than that of ductal carcinoma in situ, atypical ductalepithelial hyperplasia,intraductal papilloma,5cm breast adenosis paracancerous,organizations, the difference was statistically significant (P <0.05); while P73αmRNA in invasive breast the relative expression levels of the cancer group(0.784±0.025) was significantly higher than other groups, the differencebetween groups was statistically significant (P <0.05).14-3-3σ gene expressionrelative to the amount and organization of breast cancer grade, TNM stage andaxillary lymph node metastasis; P73α gene expression relative to the amount ofbreast cancer TNM stage and axillary lymph node metastasis.14-3-3σ andP73α expression at mRNA and protein level trend.4, invasive breast cancer tissues,14-3-3σ, p73α and p53proteinexpression in the relationship:94cases of the invasive breast cancer tissue,p53protein expression in63cases positive, accounting for67.0%, of whichp73α protein positive expression rate88.9%(56/63), and p53proteinexpression in the negative group (31/94,33.0%), p73α protein expression rate was54.8%(17/31), p73α protein expression and p53protein expressionpositively correlated, significant difference (r=0.025, p=0.000).63cases ofp53gene mutation in the breast cancer, the p73α protein expression of thenegative-positive and14-3-3σ-positive37.5%(21/56), p73α protein-negativeand14-3-3σ expression accounted for71.4%(5/7p73α expression),14-3-3σwas positively related (r=0.309, p=0.002).Conclusions1,14-3-3σ protein and mRNA levels in breast cancer with low expressionand p73α high expressions of14-3-3σ gene inactivation and p73α geneexpression closely related to the enhanced part of the breast cancer.2,14-3-3σ gene and protein in breast tissue from normal-hyperplasia-thecancer process, the positive expression tended to decrease, suggesting that14-3-3σ protein is involved in the process of breast cancer and malignanttransformation; and may be breast cancer the development of an importanttumor suppressor factor.3, P73α gene and protein expression was significantly higher in invasivecarcinoma tissue, and axillary lymph node metastasis, more common in the ER,PR-positive cases. Tip as a tumor suppressor, increased P73α expression isclosely related with the occurrence and development of breast cancer.4, Invasive breast cancer, positive P73α gene expression and p53genemutation is related to p53gene mutation in breast cancer tissue,14-3-3σpositive correlation between the expression of p73α, suggesting that p73α geneexpression was elevated when p53mutations, to replace the p53increase itsdownstream factor of14-3-3σ play a role in inhibition of cell proliferation.
