ObjectiveTo investigate the effect on mesenchymal stem cells (MSCs) of rats weredifferentiated to cardiomyocyte-like cells by different induction conditions:angiotensin II (Ang II), myocardial cell lysate induced separately andco-induction.MethodsCell adherence method for getting MSCs of rats, and then cultured MSCs’sprimary and passaged by DMEM low-sugar medium which was containing10%fetal bovine serum (FBS). The MSCs of the4th generation confirmed by CD44and CD90antibodies identificate were divided into four groups randomly whichwere0.2μmol/L Ang II induced group, myocardial cell lysate induced group,0.2μmol/L Ang II and myocardial cell lysate co-induction group, and the controlgroup. After abandoning all of the induction medium24hours later, cultured thecells continue by DMEM low-sugar medium which was containing10%FBS for4weeks. Observed the cells’ morphological change during that times. Identifieddifferentiated cells through immunocytochemical method by usingcardiac-specific troponin I (cTnI) antibody and calculated the conversion rate ofpositive cells, compared the numbers in different groups4weeks later. The statistical datas were processed and analysisde with SPSS17.0software.ResultsIn addition to the control group, other three groups of MSCs hadmorphological changes after four weeks inducing cultured. The cells’ shapewere changed from oval to spindle, polygonal and irregular shape. At the end,becoming rod-like, short rod tends to be parallet to the growth and arrangement.We also saw that there are myotube formations and protrusions around thatcells. All the groups had expressed cTnI, however, the expression was different:There were more myotube formations and higher cell conversion rate inco-induction group than the others (P<0.05).ConclusionsBoth Ang II and myocardial cell lysate can induced MSCs tocardiomyocyte-like cells, furthermore, it would have higher transformation ratewhen both reagents combined.
