ObjectiveTo detect the differention on expression of far upstream elementbinding protein1(FUBP1) in human esophageal carcinoma and itssurrounding tissue, and to explore the expression and clinicalsignificance of FUBP1in them.MethodsUsing Immunohistochemistry method the expression of FUBP1was detected in esophageal cancer from37patients, the surroundingtissue of esophageal cancer from35patients and esophagitis tissuesfrom12patients. Using Fluorescent quantitative reverse transcriptionpolymerase chain reaction(FQ-PCR)the expression of FUBP1mRNAwas measured in esophageal cancer and its surrounding tissue from32patients.ResultsThe positive rate of FUBP1with immunohistochemistry inesophageal cancer was highest among three grops (P<0.05). Therelative amount of FUBP1mRNA with FQ-PCR method in esophagealcancer tissue was higher than that of paraneoplastic tissue (P<0.05).The positive rate of FUBP1in esophageal cancer, the surroundingtissue of esophageal cancer, esophagitis tissues were81.1%(30/37),31.4%(11/35), and33.3%(3/12). Between FUBP1and clinicalparameters, including age, gender, location, length, infiltration, histological type, gross specimen type were not statistically significant(P>0.05). However, in well-differentiated, moderately differentiatedand poorly differentiated esophageal cancer, FUBP1showed agradually increasing trend. With or without lymph node metastasis, and with or without distant metastasis of tumors were significantdifference in the expression of FUBP1.The intra-coefficient of variations (CV) of FUBP1and β-actin withFQ-PCR method were0.9%–1.3%and1.0%–2.9%respectively,inter-coefficient of variations were2.9%and2.2%respectively. Therelative amount of FUBP1mRNA in tissues of esophageal carcinomaand the surrounding tissue of esophageal cancer, were0.2650±0.1356and0.1982±0.0263, Between FUBP1and clinical parameters, including age, gender were not statistically significant(P>0.05).However, in well-differentiated, moderately differentiated and poorlydifferentiated esophageal cancer, FUBP1showed a graduallyincreasing trend. With or without lymph node metastasis, and with orwithout distant metastasis of tumors were significant difference inthe expression of FUBP1.ConclusionsFUBP1plays an important role in the development ofesophageal cancer. The increased expression of FUBP1ehances thesusceptibility of canceration. The overexpression of FUBP1might becorelated with carcinogenesis, invasion, infiltration and metastasis.
