Background:In recent years,mAb is known as the most promising anti-tumor drugs. Especially specific monoclonal antibody for human DR5receptor, DR5antibody drugs are also going on the market soon, but its problems have also existed, distribution of DR5in the human body is different, the expression of DR5is also different,people think how to use its anti-tumor fuction. Naringenin is a natural flavonoids, widely present in plants and Chinese traditional medicine, Studies show that variety of pharmacological activities have been found, such as the main antioxidant, anti-aging, anti-tumor, anti-atherosclerosis, the role of DNA repairment. In this study, we chooseHepG2cells,To study the synergistic effect of mDRA-6and naringenin on liver cancer HepG2cells and its possible mechanism.Objective:To study the synergistic effect of mDRA-6and naringenin on liver cancer HepG2cells and its possible mechanism.Methods:HepG2cells were cultured with RPMI1640medium in regular condition. Cytotoxicity was examined by MTT assay. The rate of apoptosis were detected by flow cytometry with Annexin V-FITC/PI staining. DR5expression on HepG2cells surfaces was determined by flow cytometry. Casepase8expression on HepG2cells was determined by WB.Results:1.MTT analysis showed the death rates of HepG2cells was39%in the presence of10mg/L mDRA-6for12hours. When mDRA-6with naringenin at200,400,800μmol/L were used to treat HepG2cells, the death rates of HepG2cells increased to58%,79%,85%;2.Flow cytometry detection indicated the apoptosis rate of the cells was16.77%in the presence of2mg/L mDRA-6for12hours with Annexin V-FITC/PI, but the apoptosis rate of the cells increase62.66%after2mg/L mDRA-6with400μmol/L naringenin treated for12hours; 3.WB detection indicated Caspase-8were actived after mDRA-6with naringenin treated together;4.The natural expression of DR5on the surface of the HepG2cells is42%,the expression of DR5increased73%after naringenin treated.Conclusion:Naringenin can enhances the apoptotic effects of DR5monoclonal antibody on HepG2cells, The possible mechanism is that naringenin can increase DR5expression on HepG2cells surfaces.
