Objectives Human adipose-derived stem cells(hADSCs) derive from human fat tissue and has similar characteristics as bone marrow mesenchymal stem cells,and low immunogenicity.This experiment based on the Cisplatin induced Acute Kidney Injury AKI in mice,then transplanted hADSCs into mice via tail vein. Observe the effect of hADSCs to AKI, and explore the possible mechanism.Methods Twenty-four mice divided into three groups (n=8in each group), they respectively are normal group (A group), physiological saline group (B group), hADSCs group (C group).Expand the hADSCs cells in vitro and dye fluorescent DAPI.The A group not give any treatments; A group and B group injected cisplatin12.0mg/kg intraperitoneal.24hours later, the acute renal injury model was made. B group give0.2ml asepsis physiological saline. C group give0.2ml hADSCs suspension vise tail intravenous.Five days later.execute mice and remain blood and kidney specimen.exam the following indexes:①Automatic biochemical analyzer exam the level of serum creatinine and urea nitrogen;②renal histopathology under microscope;③tunel exam the apoptosis of renal tubular epithelial cells;④immunohistochemical exam the proliferating celln uclear antigen on tubular epithelial cells.⑤ELISA method exam content of TNF-1a and LI-6in kidney tissue;⑥trace fluorescent dye marked cells under Immunofluorescence microscopy.Results1.Group A blood serum creatinine is (43.68±3.60)μmol/L; the blood serum creatinine of group B and Crespectively is (120.77±15.21)μmol/L,(87.24±11.07)μmol/L, compar with group A,group B and C has difference(P<0.05).The value of group B higher than group B(P<0.05).Level of blood serum urea nitrogen of group A is (12.49±0.80)mmol/L; level of blood serum urea nitrogen of group B and C respectively is (14.74±2.24)mmol/L,(30.55±3.7)mmol/L;the value of group B and C is higher than group A (P<0.05),the value of group C is higher than group B (P<0.05).2. Group A is0, group B and group C respectively is (3.13±0.23),(2.00±0.27);the value of group B and group C higher than group A, the difference has statistics significance (P<0.01), the value of group C higher than group B(P<0.05).3.The number apoptosis of group A is (3.88±0.44);The number apoptosis of group B and group C respectively is (24.75±3.00),(18.00±2.07), compare with group A, the value of them higher than group A(P<0.05), group B higher than group C (P<0.05).4.There is no positive cells on group A.The number of group B and group C respectively is (15.63±2.71),(24.5±2.28),group B compares with group C, the value of group C higher than group B(P<0.05).5.Group A content of TNF-a is (101.65±8.52) pg/ml; The content of group B and C respectively is(203.62±25.37)pg/ml,(151.82±10.89)pg/ml, the value of them is higher than group A(P<0.05); group B higher than group C (P<0.05).The content IL-6of group A is (73.39±5.32)pg/ml; The content of B and group C group respectively is (156.57±8.69)pg/ml.(129.80±4.68)pg/ml, the value of them is higher than group A(P<0.05); group B is higher than group C (P<0.05).6.Under immune fluorescence microscope, only a small number of positive cells can be seen on kidneys frozen slice.Conclusions1.Human adipose-derived stem cells has certain effect of promote repair to cisplatin induced acute kidney injury in mice;2.The hADSCs may reduce the synthesis and release inflammatory factor TNF-1a and lL-6,cell apoptosis and upward cell proliferation to improve repair kidney function.
