Preliminary Study Of The Reversal Effects And The Inhibition Effects Of CIK On Human Cisplatin Resistant Cell Line A549/DDP

Objective:To investigate the reversal effects and the inhibition effects of CIK on human cisplatin resistant cell line A549/DDP in vitro.Methods:1.The preparation of CIK:PBMC cells were collected from20ml blood of healthy donor, it was induced by IF-γ,IL-2,CD3McAb and so on to obtain CIK cells.2.To detect the cytotoxicity of cisplatin,CIK cells and the conbination of the two above by MTT assay.Caculate the resistent index and the reversal fold.Results:1.The IC50(50%inhibiting concentration)value was1.823μg/ml and6.392μg/ml when A549and A549/DDP cells were treated with cisplatin. the two groups significantly different from controls (P<0.05). The IR=3.506.2.CIK also has an inhibition effect on both A549and A549/DDP cells. The difference between the two groups had no statistical significance (P>0.05).The IC50value was7.657×105cells/ml,7.955×105cells/ml when A549and A549/DDP cells were treated with CIK.3.Chose CIK IC10(4.130×105cells/ml) value as its non-toxic dose to exclude the influence of CIK on the A549and A549/DDP cells.Chose CIK doses at1×105cells/ml、2×105cells/ml、3×105cells/ml that bellow its IC10value treat on A549/DDP cells for44hours.Then conbined with cisplatin for4hours.. And then the IC50values decreased to1.743μg/ml、1.336μg/ml、0.883μg/ml. The difference had statistical significances (P<0.05). The Reversal Fold was3.667、4.784、7.239respectively.Conclusion:1.Cisplatin has an inhibition effect on both A549and A549/DDP cells,the effect on A549/DDP is weaker than on A549.It can be concluded that the cisplatin resistant cell line human lung asenocarcinoma (A549/DDP) is resist to cisplatin.2.CIK has an inhibition effect on both A549and A549/DDP cells. The difference between the two groups had no statistical significance (P>0.05).3.the inhibition effect of cisplatin to the A549/DDP cells is enhanced when conbined with CIK.The IR enhanced obviously with the addition of Effect/Target rate.CIK have the potential to partly reverse the drug resistance of cisplatin resistant cell line human lung asenocarcinoma (A549/DDP), the mechanisms need to be confirmed in further studies...
