Objective:To investigate the mechanism of neuroprotective effect of exogenous H2S against brain damages and cognitive deficits induced by epilepsy in rats.Methods:The epilepsy rat model was induced by intraperitoneally administration of LI-PILO, the model is successful when status epilepticus without space of seizures appear to band Ⅳ or above. Male SD rats were divided into four groups:normal control group(NC group), epilepsy group(EP group), EP+HA(Hydroxylamine,HA)group(HA group),EP+NaHS group(NaHS group). Record the delitescence attack to epilepsy Ⅳ level and above and the times of spontaneity attack of the rats. The ability of spacial learning and memory of epilepsy rats was tested by Morris water maze. The level of H2S in Hippocampus was detected by sensitive sulphur electrode method. The expression of Drebrin mRNA was observed by Real time PCR.Results:①Similar to temporal lobe epilepsy was induced by intraperitoneally injection of Lithium-Pilocarpine in rats, compared with EP and HA group. NaHS group the times of spontaneity epilepsy was decreased obviously(P<0.05).②Water maze test shows that EP,HA groups compared with the NC group the spatial learning and memory ability of rats decrease.(P<0.05). NaHS group compared with EP and HA group can improve the ability of learning and memory in EP rats,the indice datas had significant differences (P<0.05).③After the attack of SE in rats, EP group, HA and NaHS group H2S content in hippocampus are significantly lower than the NC group at three time points (3d,7d30d)(P<0.05or P<0.01); H2S content in hippocampus in NaHS group was significantly higher than in EP and HA group in3d and7d (P<0.01), but no significant difference in30d (P>0.05); HA group compared with EP group H2S content in hippocampus had no significant differences at three time points(P>0.05);④Nissl staining and quantitative analysis shows:Compared with the NC group. EP and HA group reduced significantly in the number of neuronal survival in the CA1area (P<0.01):NaHS group much more than EP group and HA group in same(P<0.01). less than the NC group, but no statistically significant differences (P>0.05): EP group compared with HA group. the number of neuronal survival in the CA1area had no significant difference (P>0.05).⑤By the compariment of each group in the expression of drebrin mRNA in hippocampus, EP, HA and NaHS group much less than the NC group in3d and7d (P<0.05or P<0.01); each group are not significant differences in the expression of drebrin mRNA in30d(P>0.05); EP group compared with HA group had no significant differences in expression of drebrin mRNA at three time points (P>0.05).Conclution:Widespread neuronal injury and cerebral cognitive dysfunction was arised after Epileptic seizures in rats; Exogenous H2S can protect neurons, up-regulate the expression of drebrin mRNA in hippocampus and moderate cerebral cognitive dysfunction during the acute Epileptic seizures phase in rats.
