Objective:To evaluate the effects on Th cells and cytokines in mouse with the avβ3intergrin antagonist SB-273005.Methods:Selected5normal mice as the blank group.Female mice were co-caged with male ones at2:1overnight to impregnate and check the following morning for vaginal plugs. The day of vaginal plug was recorded as Day1of pregnancy, that is D1. Established a animal model for mouse pregnancy. Randomly selected20mice for the normal pregnancy group,20mice as the role of drug group.Then feed integrin antagonist SB-273005by gavage, observe Th1、Th2and its secretory interleukins-IL-2、IL-10in spleen and peripheral blood by ECM and ELISA.Results:In spleen IL-2secreted by Thl decreased in preganacy and increased after drug; IL-10secreted by Th2increased in preganacy and decreased after drug (p <0.05). The data in the peripheral blood did not change significantly after treatment. The results indicated that in pregnancy Th2cytokines account for dominant and Thl cytokines account for weaknesses which the drugs can reverse the superiority.Data from flow cytometry show that in spleen Th2/Thl ratio increased in preganacy and decreased after drug (p<0.01). These results were corresponded to cytokine.Conclusion:Normal preganacy is a phenomenon of Th2shifting. Integrin antagonist SB-273005may inhibit implantation by interfering with Th2cells shift.
