Objective:To determine the major routes of buccal mucosa transport of chlorogenic acid standard substance and chlorogenic acid of throat ingate lozenge, and examine the effects of different pH, different concentration on chlorogenic acid permeation.Methods:Through the HPLC method for measuring the solubility in different pH, the oil/water distribution coefficient of chlorogenic acid and the chlorogenic acid content of throat ingate lozenge. Separate128rabbits into4groups at random averagely. Take out the buccal mucosa of the first group rabbits, use of FRANZ diffusion pool, the left buccal mucosa supplying room solution containing different concentration of chlorogenic acid standard substance which have the same pH value of7.4PBS buffer, and the right buccal mucosa FRANZ diffusion pool side is blank control group. Take1ml fluid of the accepting room after Two hours.Using HPLC method to determine the content of chlorogenic acid. The experimental method of the second group was the same as the first group, the left buccal mucosa supplying room solution containing different concentration of chlorogenic acid of throat ingate lozenge which have the same pH value of7.4PBS buffer. Take out the buccal mucosa of the third group rabbits, use of FRANZ diffusion pool, the left buccal mucosa supplying room solution containing different pH value of PBS buffer of chlorogenic acid standard substance which have the same concentration, and the right buccal mucosa FRANZ diffusion pool side is blank control group. Take fluid of the accepting room at15min,30min,60min,120min respectively and use HPLC method for measuring the content of chlorogenic acid, use the same amount of normal saline to added the fluid in the accepting room. The experimental method of the fourth group is the same as the third group, the left buccal mucosa supplying room solution containing different pH value chlorogenic acid of throat ingate lozenge PBS buffer which have the same concentration.Results:Use the one-way ANOVA, we can compare the steady state flux of chlorogenic acid standard substance which have the same pH value but different concentrations, the steady state flux of chlorogenic acid of throat ingate lozenge which have the same pH value but different concentrations respectively. We get P<0.05both, which indicates that the steady state flux of different concentrations of chlorogenic acid are different. Using the correlation to analysis the steady state flux of chlorogenic acid standard substance which have the same pH value but different concentrations. We get r=0.995, r-=0.990, P<0.05.Also use the correlation to analysis the steady state flux of chlorogenic acid of throat ingate lozenge which have the same pH value but different concentrations. We get r=0.988, r2=0.977, P<0.05. Which indicate the steady state flux of chlorogenic acid standard substance and chlorogenic acid of throat ingate lozenge are increasing with the increase of donor concentration. Use the one-way ANOVA, we can compare the permeability coefficient of chlorogenic acid standard substance which have the same chlorogenic acid concentration but different pH value, permeability coefficien of chlorogenic acid of throat ingate lozenge which have the same chlorogenic acid concentration but different pH value respectively. We get P<0.05both. Which indicate the steady state flux of chlorogenic acid among the different pH value is different. Using the correlation to analysis the permeability coefficient of chlorogenic acid standard substance which have the same concentrations but different pH value. We get r=-0.997, r2=0.994, P<0.05. Also use the correlation analysis to analysis the permeability coefficient of chlorogenic acid of throat ingate lozenge which have the same concentrations but different pH value. We get r=-0.968, r2=0.936, P<0.05. Which indicate the permeability coefficient of chlorogenic acid standard substance and chlorogenic acid of throat ingate lozenge are decreasing with the increase of donor pH value. When the pH is the same, use the correlation analysis to analysis the permeability coefficient of chlorogenic acid standard substance and distribution coefficient of chlorogenic acid. We get r=0.995, r2=0.990, P<0.05, which indicate that the two of them have a positive correlation relationship. When the pH is the same, use the correlation to analysis the permeability coefficient of chlorogenic acid of throat ingate lozenge and distribution coefficient of chlorogenic acid. We get r=0.988, r=0.977, P<0.05, which indicate that the two of them have a positive correlation relationship.Conclusion:The steady flux of chlorogenic acid standard substance and chlorogenic acid of throat ingate lozenge increases with the increase of the concentration of the drug which indicates that chlorogenic acid standard substance and chlorogenic acid of throat ingate lozenge permeates buccal mucosa via the passive diffusion way. The distribution coefficient of chlorogenic acid standard substance and chlorogenic acid of throat ingate lozenge have a linear relationship with penetration coefficient explain that chlorogenic acid standard substance and chlorogenic acid of throat ingate lozenge mainly via transcellular route penetrate cell membrane.
