Effect Of Dendritic Cells On The Pathogenesis Of Chronic Obstructive Pulmonary Disease Rats Treated With CCL Inhibitors

Objective:To study the expression of Dendritic Cells (DCs) in the lung tissue of rats with Chronic Obstructive Pulmonary Disease (COPD) and the therapeutic effect of anti-CCL20therapy on it,and discuss the possible mechanism.Methods:Wistar rats were randomly divided into three groups,normal control group(n=10) COPD model group(n=12) and CCL20inhibited model group(n=12).A control group of normal feeding,without any intervention.In the latter two groups,COPD was induced by established cigarette smoke inhalation about four weeks and intratracheal200ug/200ul of LPS solution twice(On the first day and fifteenth day).CCL20monoclonal antibody was injected intraperitoneally in CCL20inhibited model group on the first day. All animals were killed at28days.The pathomorphological changes in the lung were observed by hematoxylin-eosin staining(HE).The expressions of CD80mR、D83mRNA、D86mRNA、 and OX-62mRNA in lung tissue were observed by RT-PCR.Results:The changes of pathology of the COPD model group at28-day were the same as those of COPD patients.The expression of CD80mRNA. CD86mRNA and OX-62mRN A in the lung tissue significantly increased in COPD model group and CCL20monoclonal antibody treated group,which compared with normal control group(P<0.01);the expression of CD80mRNA、CD86mRNA and OX-62mRNA lung tissue significantly decreased in CCL20monoclonal antibody treated group which compared with COPD model group (P<0.01);the expression of CD83mRNA in the lung tissue of COPD model group were obviously lower than those in normal control group(P<0.01);the differences between COPD model group and CCL20monoclonal antibody treated group were not significant(P>0.05).Conclusion:The COPD rat models were established successfully by this combined methods.The expression of iDCs were increased in the lungs of rats with COPD. CCL20monoclonal antibody may inhibit migration of dendritic cells and lung tissue of rats with reduced expression of dendritic cells.COPD is associated with decreased numbers of mDCs.
