Experiment On Osteochondral Defects Repaired With Mosaicplasty Assisted With BMSCs As Allograft

Objective:To explore a new method, mosaicplasty assisted with BMSCs as allograft, for repair of osteochondral defects and integration of gaps.Methods:Firstly BMSCs were harvested from New Zealand White rabbit and cultured in vitro until third generation. And then osteochondral defects were made, repaired with autologous osteochondral cylinder transplantation and compound of the induced BMSCs and Pluronic F-127, which was defined as a experimental group. The pure Pluronic F-127was filled the interspace, which was taken as a control group. Finally histological and immunohistological studies were performed, and statistics analyses were also carried on.Result: Autologous osteochondral cylinder grafts in both group were not loosening^emersion and depression. In the experimental group, the tissue of the repaired interspace "death space" was becoming hyaline cartilage. Chondrocyte was disposeing regularly. And the "death spaces" were intergrated. The O’Driscoll、 Keeley and Salter histomorphology grades showed the scores were all higher in the experimental group(14.00±1.00)、(16.75±1.71)、(18.00±0.82)and(20.50±1.29)points than in the control group(7.67±0.58)、(8.00±0.82)、(8.50±0.58)and(9.00±0.82)points respectively. There were significantly differences between different treatments (F=419.302, P=0.000), different periods of healing (F=13.184, P=0.000)and exchange between different treatments and different periods of healing (F=4.982, P=0.015). There were no significantly differences between different limbs (F=0.020, P=0.890), and There were no significantly differences between others.Conclusion:Mosaicplasty assisted with BMSCs as allograft, which were used to repair osteochondral defects, can effectively integrate the "death space" in Mosaicplasty. We can get better result after this combination. And Mosaicplasty assisted with BMSCs as allograft may be a available method to repaired the osteochondral defects.
