Objective:The study was aimed to investigate the the possible mechanism ofconstraint-induced movement therapy(CIMT)and/or effective compounds group(ECG)of “BaiMai Powder” on dyskinesia caused by cerebral injury,so as to provide keyexperimental data for patients who need functional rehabilitation.Methods:Two-month-old male SD rats were used to establish the rat middlecerebral artery occlusion/reperfusion (MCAO/R)model,then rats were divided into thefollowing six groups (each group of10only): middle cerebral artery occlusion/reperfusion group(MCAO/R,intravenous injection of solvent), Extract of GinkgoBiloba positive control group(MCAO/R,intravenous injection of EGB,EGB group),constraint-induced movement therapy(MCAO/R,CIMT group), effective compoundgroup of“BaiMai Powder”(MCAO/R,intravenous injection of ECG,ECG group),constraint-induced movement therapy+effective compound group of “BaiMaiPowder”(MCAO/R,intravenous injection of ECG and CIMT,CIMT+ECG group),Sham-operated group(Sham group,intravenous injection of solvent). Intervention wastaken after building model: CIMT group (15min/time,1time/d×7d), ECG group(150mg.kg-1.d-1,1time/d×7d), EGBgroup(5.6mg.kg-1.d-1,1time/d×7d).All rats wereput to death immediately after7d. many parameters were measured: microstructure ofhistopathological morphology in focal cerebral ischemia (HE stain),numbers of Cellapoptosis(TUNEL stain),related indicators of Free radicals(SOD activity,MDAcontent, GSH content, NOS content), the expression of Bcl-2and Caspase-3(immunohistochemistry), weight(determine time: before intervention,3d,7d afterintervention),neurobehavioral study score(Level5,4points method,evaluation time:before intervention,after intervention). Experiment results were analysed by the SPSSstatistics software package (18.0). Results: All indices of the four intervention groups have improved significantlycompared with the MCAO/R group after7d intervention:1、The weight of the fourintervention groups have restored at the end of experiment.2、The neurobehavioralscore, numbers of Cell apoptosis in focal cerebral ischemia,microstructure ofhistopathological morphology in focal cerebral ischemia of the four intervention groupshave improved significantly,the improvement-degree from high to low in turn is:CIMT+ECG group> CIMT group> ECG group≈EGB group.3、The SOD activity,GSHcontent,SOD/MDA ratio of the four intervention groups have improved significantly,but the MDA、NO content have reduced significantly,the anti-free radical ability of theintervention groups from high to low in turn is: CIMT+ECG group> CIMT group>ECG group≈EGB group.4、The expression of Bcl-2in focal cerebral ischemia of theintervention groups have all improved significantly,the improvement-degree from highto low in turn is: CIMT+ECG group> CIMT group> ECG group≈EGB group,theexpression of Caspase-3in focal cerebral ischemia of the intervention groups have alldecreased significantly,the degree from low to high in turn is: CIMT+ECG group<CIMT group<ECG group≈EGB group.Conclusions:1.The MCAO/R model used in the study is valid.2. CIMT and/orECG may promote the recovery of the dyskinesia caused by cerebral injury,it showsthat mechanisms of related intervention may be as follows:improving the antioxidantscontent in brain cortex and oxidation resistance,reducing the cerebral infarctionvolume、the content of apoptosis proteins and the cell apoptosis.3.CIMT,ECG mayhave the same effect on cerebral-infarction rats, the former is better than the latter,butthe two are both better than EGB(gold standard), The curative effect of the combinationof CIMT and ECG of “BaiMai Powder”is much better,all these have proved that theintervention is effective.
