Radix Paeoniae Alba and Radix Bupleuri Chinensis are two commonly usedtraditional chinese medicine for compatibility. This work is about different compatibilityradio extract on the analgesic effect of four different mouse analgesic model.Theresults show that, when the compatibility between the two ratio of2:1, the fouranalgesic model, including acetic acid writhing, formalin method, hot water tailsreaction, heat board reaction, the analgesic effect of the extract is best, better than thesingle use of the analgesic effects of Radix Paeoniae Alba or Radix BupleuriChinensis.To explore the best extraction conditions, response surface methodology wasused to optimize the extraction conditions on the basis of the single factor experiment.The results show the best extraction conditions is as follow: extract time192min, thenumber of the extract times is1, and the solid-liquid radio is1:10.5. Under thiscondition, the analgesia rate was59.3%, with the error of the predicted value of1.82%.The extract was further studied by qualitative and quantitative analysis. thequalitative results show that: extract containing sugar, protein, saponins andflavonoids, which were also two kinds of active components of the two Chinese herbalpharmacopoeia. At the same time, the quantitative analysis of each component, theresults show that the total saponin content of0.76%, total sugar content of2.4%,3.5%of the total protein content, total flavonoid content was1.23%.Explore the Radix Paeoniae Alba and Radix Bupleuri Chinensis compatibility ratioat2:1, the mice acute toxicity test, traditional teratogenicity test and the rat chronictoxicity testing were used. The results indicate that the acute toxicity test,administered by oral gavage for14days, no deaths occurred in mice, and freedom ofmovement, eating, excretion and no abnormalities, motor coordination, convulsionsphenomenon, the coat is smooth and free of pollution. Mouse anatomy results showthat the heart, liver, spleen, lung, kidney and other organs were not abnormal change. The extract was safe and nontoxic, the maximum tolerated dose in mice is more than5g/kg. The traditional teratogenicity test results show that the Radix Paeoniae Albaand Radix Bupleuri Chinensis extract had no effect on fetal development in pregnantmice and no deformity appears. Weight by observing the placenta, fetal rat weight,fetal rat body length, tail length of fetal rats, no significant difference between thecontrol group and the experimental group. The long-term toxicity tests on rats showthat the rat were in good condition, there was no death phenomenon. Rat body weightand organ index that there is no show significant difference (p>0.05) between thehigh, medium and low-dose rats and the control group. The rat heart, liver, spleen,lung, kidney, brain, six kinds of organs had no abnormal organ damage(p>0.05).
