| Astaxanthin is a kind of food additives of strong anti-oxidation function. The natural astaxanthin in our study was extracted from the haematococcus pluvialis algae produced by United States Cyanotech Corporation, the research had investigated the antioxidant activity and stability about it, as well as the in vivo anti-inflammatory properties of astaxanthin products in which the the main raw material is the haematococcus pluvialis algae.Firstly, astaxanthin components in the Haematococcus pluvialis algae had been quantitated by HPLC method. It was showed that the astaxanthin in haematococcus pluvialis algae was1.03%.Secondly, antioxidant property of astaxanthin was evaluated in vitro. Compared with antiscorbic acid, the extract had strong scavenging effect on·OH, and O2-·. The inhibiting role on reducing capacity and scavenging effects on DPPH-was also displayed but inferior toVC.Thirdly, the stability of astaxanthin was studied. Astaxanthin is very sensitive to light, sunlight can decompose it; The temperature stability of astaxanthin is fine, the astaxanthin will be more stable below50℃, when the astaxanthin can easily be broken down beyond80℃;2%BHT help protect the astaxanthin from oxidation.Finally, to investigate the anti-inflammatory effects of astaxanthin and its mechanism, four animal models of inflammation were used.1. The model of dimethybezene-induced mouse ear edema. The results showed that the ear swelling can be inhibited by the products effectively(P<0.01).2. The model of cotton-induced rat granuloma. The results showed that product inhibition of granuloma was significant(P<0.01).3. The model of carrageena-induced rat pleurisy, the amount of pleurorrhea, the leucocyte(WBS), and PGE2in pleurorrhea, the level of SOD, MDA in serum was tested. The results showed that compared with the model group,all the parameters measured were lower in all treated groups except SOD,which activity was higher(P<0.01).4. The model of collagen Ⅱ-induced arthritis in rat.Bovine type Ⅱ collagen (CⅡ)was used to induce CIA in rats. The level of SOD, GSH-Px, TNF-a in serum examined by ELISA.Reagent, the circulating immune complexes(CIC) by polyethylene glycol(PEG) precipitation assay were detected. Parafin sections of knee joint stained with hematoxylin and eosin (HE)were prepared for histological analysis to assess joint damage. Astaxanthin products was showed the result in relieved swelling of ankle joint, improve the active of SOD and GSH-Px(P<0.01), reduced concentration of TNF-a and CIC(P<0.01). Astaxanthin products may effectively ameliorate the arthropathy in CIA rat models by improve he active of SOD and GSH-Px,reduce concentration of TNF-a, suppressed bony destruction. |
