Purpose:under the guidance of traditional chinese medicine theory, on the basis of confirming effective toxic ingredient of Aconitum Carmichalii Debx and clarify toxicity mechanism, discussing the toxic change rule in"toxic component-medicinal materials toxicity-rheum palametum toxicity-compound toxicity"of Aconitum Carmichalii Debx, to provide scientific basis on security application of clinical chinese medicine.Method:use staircase method and modified Karber"s method to determine the LD50or maximal tolerance dose of different component、medicinal materials.. rheum palametum、compound of Aconitum Carmichalii Debx, to observe primary acute toxicity symptoms spectrum following Aconitum Carmichalii Debx intoxication; use Ⅱ lead electrocardiogram to determine the electrocardiogram map of drugs, to evaluate the heart toxicity; and to evaluate the toxic change rule of Aconitum Carmichalii Debx combined with Chemical characterization of change in total alkaloid、single ester type alkaloids、double ester type alkaloids etc.Result:The content of single、double ester type alkaloids in fat-soluble alkaloid of Aconitum Carmichalii Debx is higher, which is primary toxic component, along with the decoction time increase while the medicinal materials toxicity decrease, single ester type alkaloids increase double ester type alkaloids decrease, after3hours total alkaloid goes steadily, white Radix aconite praeparata、black Radix aconite praeparata、 processed Radix aconite praeparata are not detect aconitine, the content of double ester type alkaloids in compatibility of radix aconite praeparata+dry ginger is the highest, the content of single ester type alkaloids in sini decoction is the highest.The LD50of fat-soluble alkaloid in Aconitum Carmichalii Debx is0.5mg/kg; The LD50of water extract of Radix aconite praeparata are22.4g/kg; The LD50of alconol extract of Radix aconite praeparata、white Radix aconite praeparata、black Radix aconite praeparata、processed Radix aconite praeparata are13.2g/kg、131.7g/kg、44.8g/kg、254.3g/kg; The maximal tolerance dose of white Radix aconite praeparata、black Radix aconite praeparata、processed Radix aconite praeparata of human body are533times、666times、266times;The maximal tolerance dose of water-soluble alkaloid of Aconitum Carmichalii Debx and polysaccharide of Aconitum Carmichalii Debx of human body are161times、533times.The alconol extract of heart toxicity of Radix aconite praeparata、white Radix aconite praeparata、black Radix aconite praeparata、processed Radix aconite praeparata are4.12g/kg、21.8g/kg、8.9g/kg、36.31g/kg; The water extract of heart toxicity of Radix aconite praeparata、white Radix aconite praeparata、black Radix aconite praeparata、processed Radix aconite praeparata are7.91g/kg、22.4g/kg、40.5g/kg、64.37g/kg. The heart toxicity of Radix aconite praeparata+dry ginger、Radix aconite praeparata+Glycyrrhizae、Chinese herbal Sini Decoction、dry ginger are4.7g/kg、38.4g/kg、11.6g/kg、22.22g/kg.Conclusion:The chemical composition in different component、medicinal materials、 herbal slices、compound of Aconitum Carmichalii Debx differences are significant, it expresses in pharmacological activity, especially in heart toxicity. Toxicity in component while not the same as medicinal materials、rheum palametum、compound.Combined the evaluation with chemical characterization of Acute toxicity and heart toxicity in different component、medicinal materials、rheum palametum compound of Aconitum Carmichalii Debx,which can clear the toxic change rule of Aconitum Carmichalii Debx, reference to the guidance of clinical medication.
