Studies On Chemical Constituents Of Raw Slices Of Acoitum Carmichaeli Debx

The aconite root is an indispensable and common drug in Chinese traditional medicine.The processed lateral root of Aconitum carmichaeli Debx.Now offically listed as " prepared Aconite " in the Chinese pharmacopoeia ninth edition.According to the theories of Chinese traditional medicine,the processed lateral root of Aconitum carmichaeli Debx. has porperty of hot,teasts of pungent and sweet,poisonous,and get through heart,kedney,spleen and12meridians. It has the effect of relieving pains.warming, recuperating vitality, processed lateral root of Aconitum carmichaeli Debx.has been widely used for Yang collapse,impotence coldness of extremities and uterus in the People’s Republic of China. However, raw aconite root were found be an ingredient in some prescriptions of Synopsis of Prescriptions of the Golden Chamber as well as Treatise on Febrile and Miscellaneous Diseases, raw slices of Acoitum carmichaeli Debx is raw aconite which only slice without anyother processed procedure.Accordingly investigation of the chemical constituents of raw slices of Acoitum carmichaeli Debx. were taken. To purify the extracts, MCI GEL, silica gel, Sephadex LH-20andRP-18, RP8were used in column chromatography. Then, the structures of compounds were elucidated by analyses of spectroscopic data.Thirteen compounds were isolated from the95%ethanol extraction of raw slices of Aconite root, ten of them were elucidated as hypaconitine (1), neoline (2), songorine (3), fuziline (4),8-oet-14-benzoylmesaconine(5), karakoline (6), maltol (7), β-sitosterol(8), daucosterol(9), uracil (10).Maltol were obtained from this plant for the first time.Diterpenoid alkaloids have a property of analgesic,Thus to develop a method using HPLC-ELSD for determination content of neoline which is obtained with a great quantity,Agilent1200-Alltech ELSD3300was used.The chronmatorgraphic cinditions are as following:C18column(250mm×4.6mm,5μm),the mobile phase was composed of aceionitrile-water-diethylamine (45:55:0.1),flow rate was at1.0ml/min,column temperature was30℃. the ELSD was used as detector with temperature for the detector drift tube of55℃and the nitrogen flow rate of1.5L·min-1The linear ranges of neoline was at1.944～19.44μg (R2=0.998), the average recoveries(n=6)were97.0%with RSD2.35%.The contents of neoline in raw slices of Acoitum carmichaeli Debx. is about lmg/g. This method is simple, accurate, reliable, and can be used for quality assessment.
