| Abstract:To study the effect Fas and telomeric repeat binding factor1,2on Sodium seleniteInduced human gastric cancer SGC-7901cells apoptosis and explore the mechanism of cellapoptosis.Methods:After SGC-7901cells were treated with different concentrations(0.5,2.5,5,8μmol/L)sodium selenite at24h,48h,72h and96h, Detect the expression of Fas proteinby flow cytometry. Detect the expression of TRF1ã€TRF2protein by immunohistochemi-cal SABC method. Detect the expression of TRF1ã€TRF2protein by Western Blot. Set theblank control group at the same time.Results:Flow cytometry results of Fas protein showed that: Sodium selenite improved the Fasprotein levels in SGC-7901cells; At24h, the Fas protein levels was significantly higher(P<0.05) in8μmol/L sodium selenite group than in blank control group; At48h and72h,the Fas protein levels was significantly higher (P<0.05) in5μmol/L,8μmol/L sodiumselenite group than in blank control group; At96h, the Fas protein levels was significantlyhigher (P<0.05) in2.5μmol/Lã€5μmol/Lã€8μmol/L selenite group than in blank controlgroup. Immunocytochemistry results of TRF1showed: Sodium selenite reduced the greyvalue of TRF1protein in SGC-7901cells; At24h, the grey value of TRF1protein wassignificantly lower (P<0.05) in5μmol/L,8μmol/L sodium selenite group than in blankcontrol group; At48h, the grey value of TRF1protein was significantly lower (P<0.05) in8μmol/L sodium selenite group than in blank control group; At72h, the grey value ofTRF1protein was significantly lower (P<0.05) in2.5μmol/L,5μmol/L,8μmol/L sodiumselenite group than in blank control group; At96h, the grey value of TRF1protein was significantly lower (P<0.05) in every sodium selenite group than in blank control group.Immunocytochemistry results of TRF2showed: Sodium selenite improved the grey valueof TRF2protein in SGC-7901cells; At24h and48h, the grey value of TRF2wassignificantly higher (P<0.05) in2.5μmol/Lã€5μmol/L,8μmol/L sodium selenite group thanin blank control group; At72h and96h, the grey value of TRF2was significantly higher(P<0.05) in every sodium selenite group than in blank control group.
