OBJECTIVE:This study is aim to alleviation function of Lutein on NAFLDand AS,investigate the function mechanisms of Lutein(increase the metabolism, enhanceanti-oxidize of the body, and control the action of enzyme and the express of relevant gene),and provide a new train of thought for preventing and curing NAFLDand AS.Methods: This experiment adopt1058-week-old male apoE mouses for model, which arebought from medical animal experiment centre of Peking University.After the mouses is fedwith basic feed for a week, the mouses are divided into five groups random: model groupand experiment group with low, medium and high Lutein(24mouses per group), the normalgroup with9mouses. Model group: fed with fatty feed, the experiment group with low,medium and high Lutein separately are provided with feed with25mg/kg bw,50mg/kg bw,100mg/kg bw. Lutein mixed in feed, and the normal group are fed with normal feedindstuff.(The fatty feed contains basic feed70g, lard15g, egg yolk powder10g,casein4g,cholesterol0.11g)After two months, collect blood samples from the orbits of the mouse from every group,and measure the TC and TG. Through this half-way observation, check out whether themodeling is successful, and acknowledge the preliminary result of Luteinintervention.Meanwhile, get the liver and aorta to perform biopsy by HE staining.After four months, extract blood samples from orbits from the mouse from all groupsexcept the normal one, and measure the TC and TG in serum, additionally, other relative indices are also measured. Meanwhile, get the liver and aorta to perform biopsy by HEstaining.After six months, extract blood samples from orbits from the mouse from all thesixty-four mice, and measure the content of the blood lipids, meanwhile, get the liver andaorta to perform biopsy by HE staining.Results:1.Aortic HE staining showed that lutein intervention on arterial atherosclerosis and do havesome improvement to a certain extent, especially the lutein high-dose group to improve themost significant effect;2.The body weight and serum lipid parameters of model group mice were significantlyhigher than the normal group and after intervent lutein,they were reduced;3. Abdominal fat,perirenal fat and the testis weeks fat of model group mice compared withlutein intervention coefficient were significantly decreased;4.Content of lutein in the intervention group in mouse liver homogenate lipid metabolismrelated enzyme lipoprotein lipase (LPL)and model group are significantly reduced;5.lutein intervention group of GSH and MDA results showed that leaf flavin intervention onthe body ’s antioxidant activity has significantly improved.Conclusion:High fat diet can lead to the apoE-/-mice develop atherosclerosis and hepaticsteatosis caused by atherosclerosis and non-alcoholic fatty liver, and lutein interventioncan play a significant improvement.
