Objective:To study the angiogenic role of TanshinoneⅡA on EA.hy926cells and to explore itspossible mechanism.Methods:EA.hy926cells were cultured in vitro, cell proliferation was firstly detected using MTTassay, and appropriate concentration and culture time were selected for further experiments.The cultured cells were divided into blank control group (only culture solution), low dosesof TanshinoneⅡA group, middle dose of TanshinoneⅡA group, high dose of TanshinoneⅡA group, positive control group (add basic fibroblast growth factor, bFGF). After24-hourculture, the content of vascular endothelial growth factor(VEGF) in supernatant wasdetected by enzyme-linked immunosorbent assay (ELISA) and the VEGF mRNAexpression was detected by real-time reverse transcriptase polymerase chain reaction(RT-PCR) and vascular endothelial growth factor receptor-2(VEGFR-2)protein wasdetected by immunoblotting (Western blot).Results:1.0-10mg/mL TanshinoneⅡA can promote the proliferation of EA.hy926cell after24hours, and the5.0mg/mL promote proliferative effect is most obviously, The difference ofculture time between24h and48h is not significant, consequently,the follow-up testchoose24h as the culture time. Concentration of low doses of tanshinone IIAgroup(1.0mg/mL), middle dose of TanshinoneⅡA group (2.0mg/mL), high doses ofTanshinoneⅡA group (5.0mg/mL), respectively. Compared with the control group,expression of VEGF mRNA and protein levels of VEGF and VEGFR-2in high doses ofTanshinone Ⅱ A group and positive control group cells were increased significantly(P<0.05).Conclusion:TanshinoneⅡA can promote proliferation of EA.hy926cell within a certain range, with the concentration increase, the proliferation rate was improved, but to reach a threshold thatmay inhibit cell proliferation. TanshinoneⅡA can affect the expression of VEGF/VEGFR-2in EA.hy926cells, which may be one of the molecular mechanisms of TanshinoneⅡA inthe angiogenic effects.
