Background and ObjectiveIn female primordial follicle pool is formed around birth.The majority of primordial follicles are degeneration until menarche, during the reproductive years, the continued and gradually accelerated decline will cause follicles exhausted or too less maintain dormancy and can not be activated. In mammals, the size of the primordial follicle pool and the consumption rate of primordial follicle directly determine the length of reproductive life. Our preliminary researches suggest that the expression level of898-a during ovary aging is differently and play an important role in folliculogenesis, ovarian function, according to these results here we explore in immature mice the role898-a played and the mechanisms during early folliculogenesis.MethodsUse Immunohistochemistry and Real-Time PCR to test the location and expression level of898-a in immature mice ovary. Ovary culture with monoclony antibody or recombinant protein were used to interfere the expression of898-a; Hematoxylin-Eosin staining and follicle classification counting to analyze its role in the early folliculogenesis; Real-Time PCR test the expression level of related genes of early folliculogenesis.Results898-a protein was specifically expressed on the membrane of oocytes and granulosa cells, while there was no expression of898-a in the follicular theca cell and interstitial cells. The expression level of898-a in5D、7D mice ovary were significantly up-regulated, in14D ovary the expression level was significantly down-regulated. Suppression of898-a with898-a monoclony antibody significantly reduced the initial recruitment of primordial follicle, the expression level of GDF-9、ZP1、ZP2、Cx37were significantly decreased (P<0.05), Figla、ZP3were no significant difference (P>0.05); Up-regulate898-a with898-a recombinant protein, the initial recruitment was increased while no statistical significance, the expression level of ZP2、GDF-9、Cx37were significantly increased (P<0.05),Figla、ZP1、ZP3were no significant difference (P>0.05).ConclusionIn immature mice ovary, high expression of898-a promote the initial recruitment of primordial follicle and was related to the sharply decrease of primordial follicle pool in this period.
