Objective:To analyze the expressions of delta-opioid receptor(DOR),(3-arrestinl and Bcl-2in the spleen of rats with ulcerative colitis(UC) will help to studying the effects of DOR-(3-arrestinl-Bcl-2signaling on the immuno logic pathogenesis of ulcerative colitis and the effects of Compound Kushentang on the therapeutic mechanism of ulcerative colitis.Methods:Eighty-four SD rats were divided into control group, model group, mesalazine group, Compound Kenshentang large dose group, Compound Kushentang group and Compound Kenshentang small dose group with14each randomly. All the experimental rats without control group were treated with2,4,6-trinitrobenzenesulfonic acid(TNBS) to induced ulcerative colitis. The control group and model group were administered intragastrically with normal saline, the mesalazine group were intragastrically given mesalazine solution(0.5g/L), while Compound Kushentang large dose group, Compound Kushentang group and Compound Kushentang small dose group given Compound Kushentang(0.677g/ml,0.338g/ml,0.169g/ml). The calculation of the dose of Compound Kushentang group is based on the drug dose conversion formula between human and experimental animals, in accordance with the adult dose. The experimental rats were given3ml solution everyday for fifteen days. The expressions of delta-opioid receptor, beta-arrestinl and Bcl-2from spleen tissues of rats were determined by Western blot and real-time quantitative polymerase chain reaction (RT-PCR). Results:Compared with the control group, the expression of DOR,(3-arrestinl and Bcl-2were significantly increased(P<0.05) in spleen tissues in the model group. Compared with the model group, the expression of DOR,(3-arrestinl and Bcl-2were significantly decreased(P<0.05) in spleen tissues in the treatment groups. However, there is no statistically significant differences among mesalazine group, Compound Kushentang large dose group,Compound Kushentang group and Compound Kenshentang small dose group (P>0.05).Conclusion:DOR-(3-arrestinl-Bcl-2signal transduction pathway participates in the pathogenesis of ulcerative colitis in rats. Compound Kushentang attenuates ulcerative colitis by regulateing the signal transduction pathway.
