| ObjectiveTo research the effect of GA (Gambogic acid) on cell proliferation and cellapoptosis of prostate cancer PC-3cells.MethodsProstate cancer PC-3cells were treated with different concentrations of GA for24h in vitro. The cell proliferation was assessed by CCK-8assay. Cell apoptosis wereanalyzed using AO/EB staining and FCM. The expressions of P53,Bax and Bcl-2inPC-3cells were evaluated by Western blotting, respectively.ResultsCompared with negative control cells, Gambogic Acid can not only suppressprostate cancer PC-3cells in proliferation but also induce it apoptosis in a dose-dependent manner (P<0.05). The result of CCK-8assay demonstrated that cellproliferation was significantly inhibited when the concentration of GA was more than1μM. Compared with the control group, there was more orange dense stain materialthat appeared in the nuclei by AO/EB staining. Apoptosis peaks were observed aftertreated with GA in FCM. What’s more, GA could up-regulated the expressions of P53and Bax whereas down-regulated the level of Bcl-2by Western blotting.ConclusionGambogic Acid can significantly suppress prostate cancer PC-3cells growth andalso induce it apoptosis. |
