Objective: This study replicated by STZ model of diabetes and its aerobic exercise,ginkgo polysaccharides, polysaccharide aerobic exercise combined with ginkgo threedifferent ways of intervention of exercise combined ginkgo polysaccharide weight ofeach group of diabetic rats, blood glucose, glycosylated serum protein, β cells secreteinsulin, the ability of oral glucose tolerance test and islet pathological changes andBax, Bcl-2positive expression, combined with aerobic exercise is designed to providea new diabetes drug prevention and treatment of experimental evidence.Methods:8-week-old healthy male Wistar rats (SPF grade)50,8randomly selectedas the control group (N), the remaining42as the diabetic model group,60mg/kg bodyweight by intraperitoneal injection of streptozotocin rapid urinary STZ (STZ), controlanimals intraperitoneally injected with0.1%citrate buffer. Modeled after the successof the most quiet group were randomly divided into diabetic (D), diabetes, aerobicexercise group (DS), diabetic ginkgo polysaccharide group (DM), diabetes, aerobicexercise plus drug group (DMS), each N=8only. Diabetes aerobic exercise group(DS) and diabetes aerobic exercise plus drug group (DMS) continued for6weeks offormal training, training six days a week, Sunday rest, strength20m/min, time30min.Ginkgo polysaccharide diabetic group (DM) and diabetes, aerobic exercise plus druggroup (DMS)200mg/kg body weight per day ginkgo polysaccharides fed, quiet groupof diabetic (D) fed the same volume of distilled water for6weeks. The end of theexperiment, body weight observed in each group, islet weight, fasting blood glucose(FBG), glycosylated serum protein (GSP), serum insulin (FINS), C-peptide, glucosekinase (CK), the total amount of ATP enzyme ultra, ultra-capacity Na+K+-ATPenzyme, minim Ca2+Mg2+-ATP enzyme, β-cell function index (HBCI), oral glucosetolerance test (OGTT) changes by HE staining of the islet awakened morphologicalobservation and immunohistochemical staining and semi-quantitative stereology todetect β-cell Bcl-2, Bax expression changes.Result: A one-time injection of50mg/kg body weight STZ intraperitoneally methodcan be successfully established in line with the requirements of experimental diabeticrat model. The results show that: Ginkgo polysaccharide after the intervention, body weight, GK, minim Na+K+-ATP enzyme, minim Ca2+Mg2+-ATP enzyme, c-peptidewas significantly higher (P<0.05); insulin levels are significantly higher (P<0.01);glucose, GSP decreased significantly (P<0.05); micro total amount of the enzyme ATPtended to increase, but not statistically significant. Exercise intervention, body weight,ultra-capacity Na+K+-ATP enzyme, minim Ca2+Mg2+-ATP enzyme, c-peptide wassignificantly higher (P<0.05); insulin levels are significantly higher (P<0.01); glucose,GSP decreased significantly (P<0.05); GK, minim total ATP enzyme tended toincrease, but not statistically significant. Reasonable exercise combined ginkgopolysaccharide after the intervention, GK, minim total ATP enzyme K, minimNa+K+-ATP enzyme, minim Ca2+Mg2+-ATP enzyme, insulin significantly increased(P<0.05); weight, c-peptide is very significantly higher (P<0.01); glucose, GSP issignificantly decreased (P<0.01); pathological changes in diabetic rat islets improved;bcl-2/Bax ratio increases, the islet cells in diabetic rats to repair, regeneration anddevelopment direction.Experimental Conclusion:1) Wistar rats (SPF grade) using rapid injection of50mg/kg body weightintraperitoneal chain urinary STZ (STZ) diabetic rat model can be successfullyreplicated.2) Petroleum ether defatted boiled alcohol precipitation method successfullyextract of Ginkgo biloba crude polysaccharide.3) Aerobic exercise, a combination of Ginkgo polysaccharides and weight losscan make the symptoms of diabetic rats to control; blood sugar drop.4) Aerobic exercise, a combination of both polysaccharides and ginkgo canincrease insulin secretion in diabetic rats, islet cell reserve and secretory capacity.5) HE staining biopsy shows aerobic exercise, polysaccharides and ginkgocombination allows the pathological changes of islets of diabetic rats improved.6) Bax, bcl-2expression shows aerobic exercise, a combination of bothpolysaccharides and ginkgo can make diabetic rat pancreatic β cells to repair andregeneration direction.
