Object: To observe the comparative proteomic analysis of proteins betweenhuman osteosarcoma (OS)and normal osteoblastic cell lines.Methods: OS cell (MG-63) and human normal osteoblastic cell (hFOB1.19)cultured were used as a model. Total proteins obtained in OS cells and osteoblasticcells were subjected by two-dimensional electrophoresis(2-DE). And differentproteins between OS cells and osteoblastic cells were detected by2-DE analyzingsoftware ImageScanner. Differential protein spots were characterized by matrixassisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF/TOF MS). Finally the special spot vinmentin was detected by western-blot inOS tissue of patients to verify in vivo.Result:1.The total protein profiles of two cell lines were obtain through2-DEcombined with analyzing software ImageScanner. There were73protein spotsdetected by software ImageScanner differed in2-fold between two cell lines,50spotsoverexpressed and23dawnregulated. Choose12significant differential spots betweentwo cell lines to characterize by MALDI-TOF/TOF MS. We thought vinmentin maybe OS cancer biology and potential diagnostic and treatment biomarker through theanalyze of MALDI-TOF/TOF MS and bioinformatics.2.There are many differencesbetween cell lines and human tissues. So we should carry a western-blot study on OStissue samples to further verify the results obtains from cell lines. After western-blotstudy, the expression of vinmentin in OS tissue samples(n=7) is significantup-regulated with its controls(n=7).Conclusions:1.Through comparative proteomic analysis, we can detect the differential proteins between human osteosarcoma (OS)and normal osteoblastic celllines. And these proteins may be involved in the processes of biology.2. Theexpression of vinmentin in OS tissue samples is significant up-regulated with itscontrols. So we think vinmentin may be the potential to be OS cancer biology anddiagnostic and treatment biomarker.
