Objectives: To seek the relationship between hedgehog signaling with epithelial-mesenc-hymal transition in ovarian cancer treated with cyclopamine. Methods: Molecular (E-cadherin,Vimentin) and Functional (proliferation, migration, invasiveness) levels in ovarian cancers(SKOV3, A2780, OV2008, CAOV3, C13K)treated with cyclopamine were monitored using RT-PCR, qRT-PCR, wound healing assay, transwell, Western Blot and immufluoscence. Results: In ovarian cancer cells (SKOV3,A2780,OV2008, CAOV3,C13K) treated withdifferent doses of cyclopamine, the levels of epithelial markers such as E-cadherin andB-catenin were increased, while the meschymal markers such as Vimentin and N-cadherinwere reduced; the morphology is changed and the levels of proliferation, migration and in-vasiveness were reduced,.Conclusions: Hedgehog signaling promotes epithelial-mesenchymal transition in ovariancancer, cyclopamine, a inhibitor of hedgehog signaling, may be of potential therapeuticvalue in ovarian cancer.
