Abstract/Summary:
Objective: To observe the expression of CAR (Coxsakie virus andAdenovirus receptor) in mouse uteri during implantation procedures bymeans of immunohistochemistry and real-time PCR, and assess its functions inthis biological event.Methods: Female Kunming White were divided randomly into4groups(n=7)as following: dpc(days post coitus)2.5,4.5,6.5and8.5. Both localization andquantity of CAR expression were evaluated in uteri recovered at dpc2.5,4.5,6.5,8.5by Real-time PCR and immunohistochemistry. Outcome of PCR wasassessed by2-△△CTmethod. IPP (Image Pro-Plus6.0software) was used forquantifying mean density of CAR expression in immunohistochemical sections.Statistical significance was evaluated using one-way ANOVA. Probably valuesof <0.05were considered statistically significant.Results: We found relatively weak CAR expression in mouse uteri duringimplantation window. Based on both PCR and immunohistochemistry data,highest CAR expression was shown on dpc2.5followed by down regulation ofCAR on dpc4.5and6.5(with significant difference). On dpc8.5, CARexpression rose slightly again. Also, on the section of dpc4.5, the CAR proteinstain on the non-implantation site was much deeper than that on implantation sites.Conclusion: During implantation, CAR declines both on mRNA and proteinlevels to induce the impairment of tight junction between cavity epitheliumcells. After implantation window closure, it appears again to maintainepithelium stability. CAR might play an important role during embryoimplantation procedure.
