Protective Effect Of Paidubaoshen Decoction In Renal Tubulo Interstitial Fibrosis With UUO In Rat

Objective To observe effectiveness of renal fibrosis rats with uuo treated by Paidubaoshen decoction and to get the better understanding of the possible antifibrotic mechanism of action through the experiment.Methods Through the method of uuo(Unilateral ureteral obstruction),to make rat model of renal fibrosis rats were randomly divided into3groups:rat in Chinese medicine treatment group were lavaged by Paidubaoshen decoction,rat in model control group were lavaged by Sodium Chloride,Sham operation group were lavaged by Sodium Chloride. Observed the renal fibrosis of these group through HE dyeing and Masson dyeing. Observed the experssion of TGF-β1, integrin α5β1of every group by immunohistochemical methods. To assess the experssion of chemokines(TGF-β1、integrin α5β1、E-cadherin、α-SMA) by Rt-PCR.Result (1) HE staining results did not showed significant renal interstitial fibrosis in the sham operation group.Atrophy of tubular,necrosis of epithelial cell,infiltration of lymphocyte and mononuclear cells in UUO model group.But compare with the UUO model group,level of renal interstitial fibrosis was reduced in Paidubaoshen decoction.Compared with sham operation group.(2)Masson staining results showed that area of renal interstitial fibrosis was increased in UUO model group,it suggest that model was successful (P<0.05),while Compared with UUO model group,aera of renal interstitial fibrosis was reduced(P<0.05) and level of changes pahtological of renal interstitial fibrosis was lower in UUO model group.(3) The immunohistochemical results showed that the model group, TGF β1integrin α5β1expression increased significantly, the treatment group of TGFβ1, integrin α5β1expression decreased significantly (P<0.05) compared with the model group;(4)Real-time PCR results show, the model group of TGFβ1, integrated prime α5β1and alpha-the SMA of mRNA expression was significantly increased to treat group of TGFβ1, integrated prime α5β1alpha-the SMA of the mRNA expression significantly decreased (P<0.05) compared with the model group; model groupof E-cadherin mRNA expression was significantly reduced, Tang treatment group of E-cadherin mRNA expression compared with the model group was significantly upregulated (P <0.05).Conclusion (1) Paidubaoshen decoction play the role of anti-renal fibrosis;(2) Paidubaoshen decoction can inhibit the expression of TGF-β1、integrin α5β1and renal fibrosis markers, suggesting that TGF-β1/integrin α5β1signaling pathway is the target of anti-renal fibrosis of detoxification.
