Objective:To detect circulating Toll-like receptor4and CD4~+CD25~+regulatory Tcells in patients with pancreas cancer, discuss the correlation between TLR4andTregs, provide basic theory for clinical therapy of pancreas cancer.Methods:Collect peripheral blood specimens of positive group and healthy group,extract PBMC by density gradient centrifugation, detect the expression ofTLR4mRNA by Real time PCR, and detect the percentage of Tregs by flow cytometry.Analyze the experimental data with SPSS15.0.Results:Real time PCR and flow cytometry showed that the expression ofTLR4mRNA and the percentage of Tregs in peripheral blood of patients with pancreascancer were significantly higher than healthy person, and the results from the patientswith different TNM stage were significantly different(p<0.05). Moreover, correlationanalysis showed that the expression of TLR4mRNA and the percentage of Tregs were positively correlated(r=0.673, p=0.021).Conclusion:In the immune system of patients with pancreas cancer, TLR4signaling may promote the proliferation of Treg cells and enhance its function.Regulating the TLR signal pathway may provide a new direction for tumorimmunotherapy. |
