Human umbilical cord mesenchymal stem cells (UC-MSCs) have the capacity ofself-renewal, proliferation and multilineage differentiation. UC-MSCs acceleratehematopoietic recovery, repair the damaged tissues and organs, and functionimmunosuppress. UC-MSCs have important and promising values of clinicalapplication. In order to obtain the quality stable UC-MSCs, the biology of UC-MSCsderived from the various donors should be characterized. We studied storage ofhuman umbilical cord and the culture of UC-MSCs. The biologic characteristics ofUC-MSCs derived from the various donors were compared by morphology,proliferation, multi-differentiation, and the expression of cluster of differentiation (CD)and cytokines. The culture supernatant of UC-MSCs was tested to promotehematopoietic clonization.The human umbilical cord should storage less than48h before UC-MSCs wereisolated. When storagd within48to72h, the isolation rate of UC-MSCs droppeddown to80%, and the isolated number of UC-MSCs decreased to3.83×10~4/cm, theprimary culture last10.8d. On the suitable DMEM/F12, the primary culture ofUC-MSCs was8.24d and the cell number was3.84×10~6. UC-MSCs derived fromdonor ages of20to31years shared a similar spindle-shaped morphology and grew onthe substrate. UC-MSCs could differentiate to adipocyte and osteoblast on theinducibale media. And they expressed CD90, CD105, and CD73, but did not secreteM-CSF and EPO. The supernatant of UC-MSCs culture could promote CD34+cells toproduce CFU-G, CFU-GM and CFU-M, but not CFU-E and CFU-GMEM. Theprolifieration capacity of UC-MSCs from the donors of20to23years was higher thanfrom those of28to31years. When the boy-born donors with the age of20to23years,UC-MSCs expressed higher levels of G-CSF, M-CSF, GM-CSF, IL-6, SOD2, TGF-β2,CDK6, and JAG1; hematopoietic cytokines GM-CSF, G-CSF and IL-6in thesupernatant of UC-MSCs culture were higher, which could promote CD34+cells toform more colonies.
