Aim of the study:The present study was designed to investigate antifungal activities of the ethanol-extraction from herbs combined with Fluconazole(FCZ) against clinical isolates of Candida albicans resistant to fluconazole.Methods:1.The Ethanol reflux extraction of the76herbs;2. The minimum inhibitory concentration (MIC) was determined using checkerboard microdilution assay. MIC80was determined as the lowest concentration of the drugs (alone or in combination) that inhibited fungi growth by80%, compared with that of drug-free wells;3. The time-growth curves、agar diffusion tests and in vivo experimentwere further demonstrated for synergistic effect of FRC combined with fluconazole(FCZ) against clinical isolates of Candida albicans resistant to fluconazole;4. The active extracts of FRC were obtained by solvent extraction and column chromatography of polyamide.Results:Our studys firstly found that FRC alone or in combination with fluconazole can effectively inhibit the growth of Candida albicans. The extracts from ethyl acetate and alcohol contribute to the activity of FRC. The extracts of80%of methanol、100%of methanol、acetone and10%HCl that were isolated by column chromatography of polyamide also contribute to the activity of FRC. Conclusions:Flos Rosae Chinensis(FRC) alone have certain degree of growth inhibition effect on Candida albicans. Using fluconazole combined with FRC may be an effective solution to overcome the resistance of Candida albicans..
