Recently, due to heavy driking caused the incidence of liver disease was a clear upward trend, and thus increasingly attracted attention, prevention and treatment of alcoholic liver disease has become an important issue in the medical. Soy isoflavones is a natural plant estrogen, with significant biological activity and a lot of estrogen irreplaceable advantages, and plays an important role in nutrition and health, so it received more and more attention. In this thesis, carried out the study of genistein on the preventive and protective effectes of alcoholic liver injury, the use of ethanolinduced alcohol liver injury model, were measured in serum and liver alanine aminotransferase(ALT), aspartate aminotransferase (AST) activity and malondialdehyde(MDA), superoxide dismutase (SOD), glutathione-SH(GSH), triglyceride (TG), total protein(TP) content and other indicators; mouse liver tissue sections were prepared in order to observe and analyze the morphological changes of liver cell pathology; ethanol metabolism enzyme activity, such as alcohol dehydrogenase(ADH), aniline hydroxylase(ANH) and aldehyde dehydrogenase(ALDH) were analyzed and detected, the purpose is the study of genistein on alcoholic liver injury prevention and protective effect and mechanism of the initial exploration.The results showed that:(1) genistein showed a dose-response relationship reduces the activities of ALT, AST, the content of MDA, TG and liver swelling rate,and improves the activity of SOD, the content of GSH caused by alcoholic liver injury, in addition, the prevention group was superior to the the pretection group, the two high-dose groups of every indicators both close with the control group.(2)the liver slices were observed and analyzed, we discover that the model control group liver cell show obvious lesions, such as swelling, deformation, cell cord derangement, after genistein treatment, the cytioathic effect shows significant improve, cell cord neatly arranged, and the liver biopsy pathological of prevention high-dose group is basic consistent with the control group.(3) Genistein can significant increase the activity of ALT, AST, inhibit the vitality of ANH, accelerate ethanol metabolism in the body, and reduce the concentration of ethanol in the serum, in addition, it also can promote the metabolism of ethanol to the ADH way by a small injury on the body.In summary, genistein can significant improve the alcoholic liver injury, ite mechanism may be related to the regulation of various metabolic enzyme activity. Genistein can activate the activity of ADH, ALDH in the body and inhibit the vitality of ANH, it makes that ethanol metabolizes to acetaldehyde by the ADH pathway of a lesser extent in the body injury, and reducing the metabolism of ethanol by the MEOS pathway, moreover, the improvement of ALDH activity has accelerated the metabolism rate of acetaldehyde, reduced the injury of metabolite acetaldehyde on the body. All in all, genistein through regulation of various metabolic enzyme activity, accelerate the ethanol metabolism in the body, regulate the metabolic pathways of ethanol, and reduce the extent of ethanol on the liver injury, moreover, the role of prevention is better than protective effect.
