Objective:1. To observe the expression of transforming growth factor β2(TGF-β2) in vitreous body in the patients with proliferative diabetic retinopathy (PDR), proliferative vitreoretinopathy (PVR) and the control group and to analyze its changing regulation.2. To analyze the influencing factors about the express of TGF-(32in the patients with proliferative vitreo-retinal diseases.3. To discuss the role of TGF-β2in the pathogenesis of proliferative vitreo-retinal diseasesMethods:The vitreous samples were collected during the vitrectomy from of61patients (61eyes).37patients (37eyes) with proliferative diabetic retinopathy (PDR) formed PDR group, including15males and22females. Average age of PDR group was45years old, and average fasting blood-glucose (FBG) was8.3mmol/1. Medical histories in symptomatic PDR patients were among1-31months.22patients with PDR-V were12males and10females, and average age was53.6years old, and average history was15.1months.15patients with PDR-VI were6males and9females, and average age was54.4years old, and average history was16.6months.24patients (24eyes) with proliferative vitreous retinopathy (PVR) formed PVR group, including14males and10females. Average age of PVR group was51years old. Medical histories in symptomatic PVR patients were among1-7months.12patients with PVR-C were6males and5females, and average age was50.2years old, and average history was4.6months.12patients with PVR-D were5males and7females, and average age was51.8years old, and average history was5.8months.0.1ml aqueous humor and0.2ml vitreous humor were collected, and they were kept in sterile centrifugal tubes for further research. There were8vitreous fluid samples, which were taken from donors for corneal transplantation, as normal control group. The concentrations of TGF-β2in vitreous body were detected by enzyme-linked immunosorbent assay (ELISA).Results:1. The concentrations of TGF-β2in vitreous body in the eyes of PDR, PVR and the normal control group were (22.71±2.32) ng/ml,(21.28±1.79)ng/ml,(19.00±0.62) ng/ml. there was a significant difference among the three groups (F=7.756> P=0.001). And the concentrations of TGF-β2in vitreous body of PDR group and PVR group were significantly higher than control group.And the concentrations of TGF-β2in vitreous body of PDR group and PVR group were significantly higher than control group. The concentrations of TGF-β2in vitreous body were increased in the eyes of PDR group in comparison with PVR group (t=2.573, P=0.013).2. For the patients with PDR, the concentrations of TGF-β2in vitreous body were (21.45±0.42)ng/ml、(24.85±0.36) ng/ml at V,VIstage respectively, showing a significant difference among the two stages (p=0.000).3. For the patients with PVR, the concentrations of TGF-β2in vitreous body were (19.87±0.18) ng/ml、(22.68±0.40) ng/ml at C, D stage respectively. The concentrations of TGF-β2in vitreous body were increased at the D stage in comparison with the C stage (P=0.000).4. For the patients with PDR, the concentrations of TGF-β2was (20.76±.67) ng/ml in vitreous body with PRP patients,(24.05±1.67) ng/ml without PRP.The concentrations of TGF-β2was significantly lower than the level with without PRP(t=-5.880,p=0.000)。5. For the patients with PVR, the concentrations of TGF-β2was (22.15±1.67) ng/ml in vitreous body with ocular trauma patients,(20.06±1.16) ng/ml without ocular trauma.The concentrations of TGF-β2was significantly higher than the level with without PRP (t=-0.930,p=0.359)。6. The linear regression analysis between the concentrations of TGF-β2in vitreous body in the eyes of proliferative vitreo-retinal diseases and the course of ophthalmopathy was correlation. PDR group of TGF-β2levels were positively correlated with the history of the eye(r=0.705, P=0.000). PVR group of TGF-β2levels were positively correlated with the history of the eye(r=0.934,.P=0.000).7. For the patients with proliferative vitreo-retinal diseases, there was no correlation between the sex, the age and TGF-β2concentration in vitreous respectively (r=-0.207, β=0.08; r=0.016, β=0.89). There was no correlation between fasting blood glucose (FBG), duration of DM and TGF-β2concentration in vitreous respectively (r=-0.159, β=0.33; r=0.012, β=0.94).Conclusion:1. Intravitreous TGF-β2level in the patients with proliferative vitreo-retinal diseases was higher than the control group. The more severe degree of fundus pathological changes was, the higher level of intravitreous TGF-P2was in the patients.2. TGF-β2may play an important role in the pathogenesis of proliferative vitreo-retinal diseases.
