Objective and MethodsMyelosuppression is one of the common side—effect for cancer patients when they receiving radiotherapy and chemotherapy. The number of leucocyte, erythrocyte, and platelet decrease severely, what’s more, excess myelosuppression may also cause infection, imflamation and bleeding,and the course of radio-and chemotherapy may be hampered, which ale limiting factors for improving the curative effect of radio-and chemotherapy. Therefore, the prevention of myelosuppression has been the. focus of many experimental studies. From TCM perspective, the pathogenesis of myelosuppression is due to the asthenia of spleen and kidney, which are the sources of congenital-Qi,blood,and kidney-Yang. Shengxuewan(SXW) is a Compound Preparation which composed of Hairy Antler, Amur Corktree Bark, Common Yam Rhizome, Largehead Atractylodes Rhizome, mulberry twig, hyacinth bean.germinated rice-grain, Dried Human Placenta and other traditional chinese medicines, those medicine are of great value in invigorating congenital-Qi and reinforcing kidney-yang. Previous study had demonstrated that this compound can efficaciously enhance body immunity. To evaluate the effect of Shengxuewan on bone marrow hematopoiesis,in our study,the survival proliferation,differentiation,apoptosis of the hematopoietic cells and the effect of Shengxuewan on bone marrow hematopoiesis was investigated, the mechanism and the characteristics of Shengxuewan on hematopoietic regulation was also explored. So as to provide an experimental basis for Shengxuewan’s clinic application.Result1.High dose Shengxuewan Can promote the recovery of granulocyte, erythrocyte, Hb,Plt and BMC. The quantities of Ret are improved in Shengxuewan group and Control group(p<0.05). The morphous of Ret in shengxuewan group are more close to normal ones.2. The two groups can effectively improve the yeilds of HSC in every series and have no difference between them(p>0.05).3. Shengxuewan and Control group have no difference (p>0.05) in releasing the stasis of the Go/Gj phase in BMC and impose them into cell cycle.The cell ratio of BMC in Go/G1phase was significantly decreased after Shengxuewan’s administration, whereas the ratio of BMC in S phase and G2/M phase was significantly increased, implied that Shengxuewan Can active BMC from stasis into cell cycling, accelerate the synthesie of DNA and the mitosis of BMC,promote the proliferation of hematopoietic cells, which will contribute to the recovery of the hematopoietic function.ConelusionWe first studied Shengxuewan’s function on hematopoietic regulation and explored it’s mechanism by evaluated the survival, proliferation, cell cycle, hematopoietic growth factor(GFM-CSF、TPO), the result of our study demonstrated that the myelosuppression which induced by irradiation and chemotherapy agent was significantly relieved after Shengxuewan’s administration, Shengxuewan may play an important role in the rehabilitation of hematopoietic function in myelosuppressed mice.
