Geniposide is one of the most important active compounds in Gardenia jasminoides Ellis, and showed to possess detoxification, anti-tumor and anti-thrombotic activities. Geniposide was normally obtained by chromatographic methods, which was disadvantage of complicated process, low yield and time comsumering. Seeking for a simple and effective large preparation of giniposide promote us to investigate new separation. A selective extraction, simple and effective method, has been established for preparation of geniposide from Gardenia fructus fruit in our recent works. Geniposide was prepared in large-scale from Gardenia fructus by selective extraction method with polymers such as sodium carboxymethyl cellulose (CMC), polyethylene glycol (PEG), polyacrylamide (PAM), in stationary phases. The selective extraction of geniposide from mixture was due to different intermolecular interactions among the polymer and compounds. Separation efficiency of two types of extraction (normal phase extraction and reversed phase extraction) was different obviously. As normal extraction, a residue of extraction from Gardenia fructus fruit was dissolved in aqueous solution of polymer, and then the aqueous solution was extracted with an organic solvent to give geniposide. In reversed extraction, the residue was dissolved in organic solvent, and then was extracted with aqueous solution of polymer. Partition behavior of geniposide between the two-phase system was investigated. Effects of polymers, concentration of polymer, the extraction repeats on separation were investigated. Investigating results gave optimized conditions of reversed phase as following.(1) The residue was dissolved in n-butanol, then was extracted with aqueous solution of CMC solution (0.5%, weight) for3times to give geniposide with a purity of85.0%.(2) The residue was dissolved in n-butanol, then was extracted with aqueous solution of PEG-6000(5.0%, weight) for3times to give geniposide with a purity of82.3%.(3) The residue was dissolved in n-butanol, and then was extracted with aqueous solution of PAM solution (2.0%, weight) for2times gave geniposide with a purity of87.1%. The investigating results showed that sodium carboxy methyl cellulose, polyethylene glycol, polyacrylamide can selectively adsorb impurities of residue, to give geniposide with better purity. Preparation of geniposide from Gardenia fructus fruit with selective extraction of polymer was feasible.The process was simple and efficient, was proved to be useful for industrial application of geniposide preparation.
