Preliminary Study On The Toxicity Of Nitrozuril And Its Efficacy Against E.Tenella In Chicken

Nitrozurol, a triazinetrione derivative, is synthesized by us now. In this article, primary study on the toxicity and pharmacodynamics effects of Nitrozuril was performed to provide theoretical foundation for its clinical application in veterinary practice.1Determination of Nitrozuril by HPLCWe determined the UVmaximum absorb chromatograms of Nitrozuril by UV and content of Nitrozuril ormalization method. The result indicated wavelength was set at300nm, The content of Nitrozuril is99.12%; the within-day coefficient of variation of2.5μg/mL; intramural was1.06%and the between-day was1.27%. The method was simple and accurate enough to determine content of Nitrozuril.2Study on the general toxicity of NitrozurilAcute toxic effect and accumulative toxic effect of Nitrozuril were detected by modified Kaber method and accumulation coefficient method respectively. The mice took orally with Nitrozuril subsequently for30d to evaluate the sub-chronic toxicity of Nitrozuril. It was tested out that the half death quantity (LD50) was768mg/kg and the95%confidence interval was644～916mg/kg. The accumulative coefficient (K) was4.26. In the subchronic toxicity test, the high groups of the organ coefficients in liver were significant lower than the control. The results of blood routine examination and biochemical index of blood in mice indicated that UA, TC and TG of the high groups were significant lower than the control and RBC and Cr were opposite. The pathohistology checks indicated that the liver was slightly hydropic degeneration of high groups. The results showed that the Nitrozuril had some toxicity when administered with a high dosage for a long time.3Study on The inherent Toxicity of NitrozurilMicronucleus test, sperm structural test and Ames test to Nitrozuril were carried out in KM mice to evaluate its mutagenic toxicity. The results of the sperm structural test and micronucleus test are negative and the Ames test is masccline. Nitrozuril has mutagenic toxicity but no aberration to KM mice.4PreventionTrials of Nitrozuril against coccidiosis in chickensWith1day old healthy chicken as the tested animals and the14day old chicken were artificially infected by E. tenellar. Then the Nitrozuril with1,2,3,4,5,6mg/kg were added to chicken. The results showed that the anticoccidial index (ACI) of Eimeria tenella of Nitrozuril were187.4,206.2,205.9,202.7,202.4and208.1. Additive content of Nitrozuril at the dosage of lmg/kg can see little bloody and Cecal lesion. This revealed that Nitrozuril is an efficient and safe new triazines compound. At the dosage of3mg/kg is suitable. Nitrozuril would expect to develop into a new anticoccidial drugs.
