Background and Objective: In previous study, we use to test14112human genes cDNA chip,20nasopharyngeal carcinaoma patients tissue samplesand15chronic inflammation of the nasopharynx tissue samples expressionprofile analysis to find in nasopharyngeal carcinoma organization have obviousdifference of gene expression. The experimental objective is:1. choosing relatedgene complement component receptor2(CR2)and MLX interacting protein(MLXIP), in50case of nasopharyngeal carcinaoma tissue to RT-PCR, to seeif the result and gene chip to express is the same;2. choosing radiation sensitiverelated gene zinc finger protein608(ZNF608) and colony stimulating factor1receptor(CSF1R), in the case of nasopharyngeal carcinaoma tissue to RT-PCR,to see if the result and gene chip to express is the same;3. analysising theexpression of CR2, MLXIP, ZNF608, and CSF1R in nasopharyngeal carcinaomaand organization of clinical relevance.Methods: In our study, RT-PCR was used to validate results of2groups in50and17NPC tissues. To validate the expression of CR2, MLXIP, ZNF608andCSF1R in nasopharyngeal carcinoma and analyze clinical significance. Results:1. In order to further research, we choose MLXIP and CR2in another50NPC tissues to proceed RT-PCR. Through comparing the other3chronicinflammation of the profile, CR2was lower expression in NPC patients andMLXIP was over expression in NPC patients.In previous sduty we have alreadyknown the expression profile analysis results of chip,nine genes(q<0.01) at leastin17NPC tissues are2times differentially expressed genes,8of themdown(TAOK3, SLC16A2, PRB4, AMY2B, B3GALT4, MSMB, RPS27, CR2),a raised gene(MLXIP). The result of RT-PCR is the same with cDNAmicroarray.2. The expression of CR2and MLXIP was not correlated to the age, sex,clinical staging and, T staging and cervical lymph node metastases(P>0.05).3. The result of RT-PCR show that ZNF608and CSF1R were overexpression in radiation resitant NPC patients and were lower expression inradiation sensitive NPC patients. In previous sduty, ZNF608and CSF1R thatresult in chip were over expression in radiation resitant NPC patients and werelower expression in radiation sensitive NPC patients. Their expressiondifferences multiples were4.10times and1.89times. This is the same withcDNA microarray.4. The expression of ZNF608and CSF1R was not correlated to theage,clinical staging, T staging and cervical lymph node metastases(P>0.05).Conclusions:1. Comparing with inflammation tissue, CR2in41case(82%) ofnasopharyngeal carcinaoma tissue is lower expression, MLXIP in42case(84%)of nasopharyngeal carcinaoma tissue is was over expression. This is the samewith cDNA microarray. CR2and MLXIP are obvious differences expression in nasopharyngeal carcinoma tissues. CR2and MLXIP would have effect in thediagnosis of nasopharyngeal carcinoma.2. The expression of CR2and MLXIP was not correlated to the age, sex,clinical staging and, T staging and cervical lymph node metastases.3. ZNF608and CSF1R were over expression in radiation resitant NPCpatients and were lower expression in radiation sensitive NPC patients. This isthe same with cDNA microarray. ZNF608and CSF1R would be a target whichmay forecast the radiosensitivity in nasopharyngeal carcinoma patients.4. The expression of ZNF608and CSF1R was not correlated to the age,clinical staging, T staging and cervical lymph node metastases.
