Objective:To evaluate the association of G196A single nucleotidepolymorphism(SNP)in brain-derived neurotrophic factor(BDNF)gene withAlzheimer’s disease(AD)in Chinese population.Methods: G196A SNP in BDNF gene was detected by polymerase chainreaction(PCR-RFLP)technique in58patients with AD and52health agedcontrols.we analysed the distribution of BDNF genotypes and alleles inChinese population.we analysed the association of genotypes with gender aswell as the severity of the disease.The risk of G196A single nucleotidepolymorphism for AD was analysed by the he non-conditional logisticregression model.Results:1、There was no significant difference in age (P=0.964) andgender (P=0.976)in AD group and control group,The genotyping distribution in both patients and controls was within Hardy–Weinberg equilibrium.Nosignificant differences were demonstrated in G196A in BDNF genotype or allelefrequencies between AD patients and controls(χ2=0.013,P=0.994>0.05,χ2=0.013,P=0.910>0.05).When AD and control groups were stratified bygender,no significant differences were observed in gender subsets(P>0.05).2、The risk of G196A single nucleotide polymorphism for AD:comparedwith the genotype GG,there was no statistical association of the risk ofindividuals who carried genotype AA for AD(OR=0.942,95%CI:0.333~2.670),there was no statistical association of the risk of individuals who carriedgenotype AG for AD(OR=0.980,95%CI:0.399~2.404),there was also nostatistical association of the risk of individuals who carried A allele for AD(OR=1.034,95%CI:0.446~2.397).3、There was no significant difference in genotypes of G196A in BDNFand the severity of AD.Conclusions:1、The distribution genotypes and alleles of G196A in BDNFin Chinese population may not be associated with AD;The SNP of G/196A inBDNF may not be the risk for AD.Both of them needs to be confirmed byexpanding the sample size and studying in multi centers.
