Gly482Ser Variability Analysis Of Peroxisome Proliferator-activated Receptor-r Coactivator-1A (Pgc-1A) Gene In Chinese Han Population With Gestational Diabetes Mellitus

Introduction Gestational diabetes mellitus (GDM) and regular type2diabetes mellitus (T2DM) share many common features, including impaired beta-cell function, insulin resistance as well as the similar genetic background. Accumulating literature had suggested that the polymorphism of peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α) gene was associated with the pathophysiology of T2DM. Nevertheless, compared with T2DM, genetic studies on GDM are relatively scarce.Objective Our study aimed to explore the relationship between the Gly482Ser single nucleotide polymorphism (SNP) of PGC-1α gene and GDM in the Southern Chinese Han population.Methods This is a case-control study involving234subjects,84of whom were GDM and150were non-diabetic pregnant women. Gly482Ser polymorphism of PGC-la gene was analyzed using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). All of the data were analyzed by SPSS13.0. Results In comparison with the controls, the patients with GDM had significantly higher progestational age (p<0.001), progestational BMI (P=0.019) and weight gain during pregnancy (P=0.029). No significant difference was found in allele distribution and genotype frequency of PGC-la SNP between the GDM and controls (p>0.05). However, there was significant difference in weight gain during pregnancy and genotype only within the control group (p=0.05, by one-way ANOVA).Conclusions Base on our research data, we concluded that the Gly482Ser SNP of PGC-la gene is not associated with the occurrence of GDM in the Southern Chinese Chinese population.
