Objectives:1.To determine the preparation process and main componentscontent of Effective Fraction of Polyphenols of Pomegranate Peel.2.To establish thequality standards and the fingerprint of the Effective Fraction of Polyphenols ofPomegranate Peel through quality analysis, and to discuss the stability of this part.Methods:1.With the total polyphenols content of the Effective Fraction as an index,We selected the preparation process of the Effective Fraction of Polyphenols ofPomegranate Peel by monofactor test.2.We determined the contents of thepunicalagin, gallic acid, ellagic acid in the Effective Fraction of Polyphenols ofPomegranate Peel through the application of HPLC,At the same time,We establishedthe quality standards and fingerprint of the Effective Fraction of Polyphenols ofPomegranate Peel through the investigation of ten batches of extract, and tooksimilarity analysis.3.The stability of the Effective Fraction of Polyphenols ofPomegranate Peel was confirmed through the observation of the variation ofappearance character and essential component contents under the condition of hightemperature, high humidity, strong light irradiation test.Results:1.The preparationprocess of the Effective Fraction of Polyphenols of Pomegranate Peel was asfollows:First,granatum was crushed through80mesh sieve and extracted by60%ethanol for0.5hour at room temperature with the ratio of material to solvent as1:24.Second,the filtrate was concentrated under vacuum to the right amount anddiluted with water containing20.8mg crude drug/mL.Third, The optimum absorptionconditions of macroporous resin HPD500were sample of2times column volume,pHvalue2, feeding rate2BV/h, adsorpting time12h. And the optimum desorptionconditions were determined as7times column volume of distilled water and8timescolumn volume of50%ethanol,collecting the ethanol eluate, obtained byconcentrating under vacuum and freeze drying.2.The contents of the ellagic acid, gallic acid, punicalagin in the Effective Fraction of Polyphenols of Pomegranate Peeldetermined by HPLC were20.47%,0.89%, and40.70%. The precision, stability, andrepeatability of the HPLC fingerprint of the Effective Fraction of Polyphenols ofPomegranate Peel were proved by scientific test, and the similaritis of the ten batcheswere all above0.990.3.The Effective Fraction of Polyphenols of Pomegranate Peelhad the ability of moisture absorption and was sensitive to strong light, which wasmainly reflected in the reduction of content of the effective components, therefore,tips should be protected from light, cool, dry place.Conclusions:1.After the purifiedby macroporous,the total polyphenols content of the the Effective Fraction ofPolyphenols of Pomegranate Peel is significantly improved, and the method is simple,efficient and low toxicity, which provides a strong technical guarantee for the furtherproduction of amplification.2.The fingerprinting and content determination method byHPLC can control the quality validly to the production of Effective Fraction ofPolyphenols of Pomegranate Peel.3.By examining the factors affecting the stability ofEffective Fraction of Polyphenols of Pomegranate Peel, the study provides a referencerange for the further use, storage, transportation and preparation of the EffectiveFraction of Polyphenols of Pomegranate Peel.
