Centipeda minima is an annual herbaceous plant of Composita-e,whi ch is recorded in Chinese Pharmacopoeia(2010I).It common-ly used as a treatment for anemofrigid headache, cough, nasal tampon, nasosinusiti s, rheumatism arthralgia, Cameroon fever, venomous snake bite and so on. Recent pharmacological study has proved it has a significant anti-t umor activity.The early work found Brevilin,ll,13-dihydrohelenalin and t araxasterol has goodantitumor activity, but the material basis of anti-turn or effe-ct was not very clear.In order to clarify its material foundation for anti-tumor effect, we extracted Centipeda minima by system solvent approach.Finally seperated the extracts into ethyl acetate fraction and n-Butanol fraction. Adopting silica gel column chromatography,gel per-meation chromatography, MCI-GEL CHP20P column chromatography and other methods, we isolated and purified the chemical composition of the antitumor active fraction, t he ethyl acetate fraction, got11chemical compounds. Those compound s mentioned above have been identified by NMR,MS and other spectral technologies are β-sitosterol, Dl,5a,8a-epidioxy-(22E,20,24R)-ergosta-6,22-dien-3β-o1,D2,Taraxasteryl acetate,D3,Lupeol,D4,Brevilin, D5,Spinastero1,D6,β-sitosterylp-alm itate,D7,3-methoxyquercetin,D8,Quercetin,D9,10-Hyd roxy-8,9-dioxyisopropylidene-thymol, D10,Centipedap-henol,D11. We screened the anti-tumor active constituents from the compounds above by MTT assay and4of them showed significant anti-tumor acti-vity. They were Lupeol,D4,Brevilin,D5,3-methoxyq-uercetin,D8,Quercetin, D9.And their IC50were53.9μg-mL-1.7.6μg-mL-1、47.5μg·mL-1,26.5μg·m L-1respectively. Also,3compounds possessing certain antitumor activity were screened. They are5a,8a-epidioxy-(22E,20S,24R)ergosta-6,22-dien-3β-ol,D2,10-Hydroxy-8,9-dioxyisopropylidene-thymol,D10and Centipedaphe n-ol,D11.And their IC50were147.1μg·mL-1、176.3μg·mL-1、184.5μg·mL-1. This dissertation provided a scientific basis so as to further clarify the material basis of the antitumor effect of Centipeda minima. It also enri ched Centipeda minima active ingredients’research content.
