Study On The Sedative And Hypnotic Effective Substances And Quality Standards Of Valeriana Of Officinalis L.

Valeriana officinal is L. belongs to the family of Valerianace and Valerian L., it is a herbal traditional Chinese medicine which have sedative and antispasmodic effects,using for disquiet insomnia、spasm、mania、hysteria and bruises.It had long been as a sedative in our people. It main has volatile oil、iridoid. sesquiterpenes、flavonoids and alkaloids five types of chemical composition. Valerian volatile oil had obvious sedative and hypnotic effects, but other substances in valerian which have sedative and hypnotic effects are not very clear. We have studied the sedative effective substances in valerian, including pharmacological selection of the effective extracts、serum pharmacochemistry and quality standards.Through the modern pharmacology methods,it have studied the sedative and hypnotic effects experiment of Valerian volatile oil and water extract on the pharmacology and the acute toxicity experiment of Valerian volatile oil.The results showed that Valerian volatile oil had obvious sedative and hypnotic effects, but it had toxicity to mice; the water extract which was extracted after Valerian volatile oil had sedative and hypnotic effects weakly. Therefore, the test result verified that Valerian volatile oil was the primary bioactive component; the result of the acute toxicity experiment could deliver the proof for selecting dose in the test of cooperative effect with pentobarbital sodium.Valerian volatile oil was abstracted from Valerian by CO2supercritical fluid extraction method and steam distillation extraction method. Their components and contents were analyzed by GC-MS. The results showed that CO2supercritical fluid extraction method was more satisfactory. So it could deliver the proof of extraction technology reform for the herbal medicine effective material part.Through the serum pharmacochemistry method in Chinese medicine and GC-MS technology,the Valerian effective material part which entered the blood were analyzed. The results showed that the ingredients in the blood were Bornyl acetate、Myrtenyl acetate、3-Cyclohexene-1-methanol,. alpha.,. alpha.,4-trimethyl-, acetate. Caryophyllene、1H-Cyclopropa [a]naphthalene,1a,2,4,5,6,7,7a,7b-Octahydro-1,1,7,7a-tetramethyl-,[(laR-(la. alpha.,7. alpha.,7a. al pha.,7b. alpha.)]-、cis-Z-. alpha.,-Bisabolene epoxide、Cedran-diol,8S,14-、1,6,10-Dodecatrien-3-ol,3,7,11-trimethyl-,[S-(Z)]-2(1H) Naphthalenone,3,5,6,7,8,8a-hexahydro-4,8a-Dimethyl-6-(1-methylethenyl)-、4,6,6-Trimethyl-2-(3-methylbuta-1,3-dienyl)-3-oxatricyclo[5.1.0.0(2,4)]octane. The study results have the important meaning to the material basis of the valerian’s sedative and hypnotic effects.Reference to the standardization of Chinese herbal medicine quality standards which were contained in the2010version of "Chinese Pharmacopoeia",and though the modern analysis method, moisture、totalasth、acidinsoluble ash、extract、TLC identification、 Valerian acid content of Valerian were examined systematically. The results showed that a comprehensive quality standards were established, it can effectively control the quality.The Valerian quality standards will be expected to provide a reference for the reprinted "Chinese Pharmacopoeia".
